大黄附子汤对重症急性胰腺炎大鼠细胞因子的影响

【摘要】 目的 :研究中药大黄附子汤对重症急性胰腺炎 ( SAP)大鼠细胞因子的影响。方法 :采用胆胰管内逆行注入质量分数为 1.5 %的去氧胆酸钠 ( 1ml/ mg)方法建立大鼠 SAP模型。选用清洁级健康雄性 SD大鼠共6 0只 ,体质量 2 6 0~ 30 0 g,随机分成中药组、模型组和假手术对照组。检测不同时间段各组血清淀粉酶、肿瘤坏死因子 α( TNFα)、白细胞介素 1β( IL 1β)及 IL 18水平 ,探讨大黄附子汤与细胞因子释放间的关系。结果 :模型组血清淀粉酶与 TNFα、IL 1β及 IL 18水平变化呈正相关 ,血清淀粉酶、TNFα、IL 1β及IL 18明显高于假手术对照组 ( P均 <0 .0 1) ;中药组血清淀粉酶、TNFα、IL 1β及 IL 18水平与模型组相比均明显降低 ( P均 <0 .0 1)。结论 :中药大黄附子汤对 SAP的防治作用可能在于使大鼠血清 TNFα及 IL 1β、IL 18水平下调

【Abstract】 Objective: To study the effect of traditional Chinese medicine Dahuang Fuzi decoction(DHFZD,大黄附子汤) on the expression of cytokines in severe acute pancreatitis (SAP) rats. Methods: The SAP models of rats were constructed by inversively pouring of 1 5% sodium deoxycholate (1 ml/mg) through cholecystpancreatic duct in rats. Sixty male SD rats (260300 g) were divided into the control group, model group and DHFZD group randomly. The levels of amylase, tumor necrosis factorα(TNFα), interleukin1β (IL1β) and IL18 in serum were detected at different interval to explore the effect of DHFZD on cytokines expression. Results: Compared with the control group, the amylase, TNFα, IL1β and IL18 levels in serum in the model group were increased significantly and a positive correlation between the expression of amylase and TNFα, IL1β, IL18 was observed(all P <0 01). After the treatment with DHFZD , the levels of amylase, TNFα, IL1β and IL18 were decreased significantly compared with those of the model group(all P <0 01). Conclusion: DHFZD has prevention and treatment effect on SAP and downregulating the levels of TNFα,IL1β and IL18 in serum may be the mechanism.
