应用Sysmex SE-9500血细胞分析仪测定干细胞的初步研究

【摘要】 目的 用BD FACSCalibur流式细胞仪评价SysmexSE 95 0 0血细胞分析仪的干细胞检测程序 (SCMP)测定干细胞的能力 ,以及探讨用SysmexSE 95 0 0血细胞分析仪测定干细胞的影响因素。方法 采用 31例外周血和脐带血标本 ,分别用SysmexSE 95 0 0血细胞分析仪测定其HPC(造血干细胞 )和用BD FACSCalibur流式细胞仪测定CD34+细胞值 ,分析两者的相关性 ,用流式细胞仪初步评价SysmexSE 95 0 0血细胞分析仪测定干细胞的能力 ;分别用肝素、EDTA抗凝 ,用RPMI 16 4 0保存的干细胞在不同的时间点测定以探讨标本储存时间对测定结果的影响及筛选理想的抗凝剂。结果 两种测定方法的相关性良好 ,相关系数r=0 80 95 (n =31) ,经t检验P <0 0 5。EDTA抗凝的标本干细胞数较为稳定 ,标本在 2~ 3小时后测定值开始减少。结论 SCMP可望在外周血干细胞移植中发挥作用 ,应使用EDTA抗凝 ,标本放置不应太长。

【Abstract】 Objective To evaluate credibility of determinating stem cell of Stem Cell Monitor Program (SCMP) of Sysmex SE-9500 compared with the determination Results of BD-FACSCalibur Flow Cytometry. Methods 31 samples of peripheral blood and umbilical cord blood were determinated by Sysmex SE-9500 and BD-FACSCalibur at the same time, relativity analysis were made between determination values of two methods, a preliminary assessment was made to the capability of determinating stem cell with Sysmex SE-9500. Heparin and EDTA were used to screen the optimal coagulant. Stem cell stored in RPMI-1640 were determinated at different time point to investigate the influence of store time on the determinating results of stem cell. Results Good correlation(r=0.809 5,n=31)between HPC and CD34-positive cell counts, stable results were obtained with EDTA coagulant, and deteminating results decreased after 2~3 hours. Conclusion SCMP might play an importantrole in the stem cell transplantation, EDTA should be used as coagulant and samples should not be stored too long prior to analysis.
