重组猪生长激素对猪脂肪组织GH-R、IGF-1、IGF-R和Leptin基因表达的影响

【摘要】 选用体质量 5 0 kg左右的去势长白×大约克公猪 16头 ,随机分为试验组和对照组 ,试验组猪每日肌肉注射重组猪生长激素 (rp GH) 4 m g/头 ,2 8d后屠宰采集背部皮下脂肪 ,用 RT- PCR方法 ,以 18S r RNA作内标 ,定量分析脂肪组织中生长激素受体 (GH- R)、胰岛素样生长因子 - 1(IGF- 1)、胰岛素样生长因子 型受体 (IGF- R)和瘦蛋白(L eptin)的 m RNA丰度。结果显示 ,试验猪脂肪细胞 GH- R m RNA丰度比对照猪增加了 5 0 .9% (P<0 .0 5 ) ,IGF- 1m RNA丰度增加 2 7.8% (P<0 .0 5 ) ,IGF- R m RNA丰度增加 4 8.1% (P<0 .0 1) ,而 L eptin m RNA丰度则比对照猪减少 2 8.2 % (P<0 .0 5 )。结果提示 ,生长激素不仅可以直接调节脂肪组织 ,还可能通过脂肪 GH- R介导产生 IGF- 以旁 (自 )分泌形式调节脂肪组织的代谢

【Abstract】 Sixteen castrated male Large White × Landrace pigs(50.0±3.5) kg were allotted into treatment and control group.The treatment group was injected intramuscularly with recombinant porcine growth hormone(rpGH,4 mg·d-1) and the control group with vehicle for 28 days.Subcutaneous adipose tissues were sampled and the total RNA was extracted.The abundance of growth hormone receptor(GH-R),insulin-like growth factor 1(IGF-1),insulin-like growth factor Ⅰ receptor(IGF-ⅠR) and leptin mRNA were measured by RT-PCR with 18S rRNA as internal standard.The results indicated that the relative abundance of GH-R,IGF-1 and IGFⅠR mRNA in adipose tissue increased by 50.9%(P<0.05),27.8%(P<0.05) and 48.1%(P<0.01) respectively by GH administration,while the relative abundance of leptin mRNA in adipose tissue were decreased by 28.2%(P<0.05) in treatment group.These results suggest that rpGH could not only regulate adipose tissue directly,but also enhanced IGF-1 secretion in adipose tissue,and that IGF-1 could act locally via the IGF-ⅠR in an auto/paracrine manner.
