IL-1β、TNF-α、TGF-β1与缺血性脑损害相关性研究

【摘要】 目的 探索白细胞介素 -1 β(IL-1 β)、肿瘤坏死因子 -α(TNF-α)及转化生长因子 -β1 (TGF-β1 )在缺血性脑卒中发病过程中的作用及相互关系。方法 采用放射免疫分析法和 ELISA双抗体夹心法检测 46例急性期脑梗死患者、46例颈动脉粥样硬化症患者及 2 2名健康者血清上述 3种细胞因子水平。结果 急性期脑梗死组血清 3种细胞因子水平均较颈动脉粥样硬化组明显升高 ,颈动脉粥样硬化组较健康对照组升高 ;颈动脉粥样硬化组和急性期脑梗死组患者血清 TNF-α与 TGF-β1水平均呈直线正相关。结论  IL-1β、TNF-α及 TGF-β1表达增强是急性期脑缺血性损伤反应 ,且在颈动脉粥样硬化期已异常升高 ,提示它们可能参与缺血性脑卒中的发病过程。

【Abstract】 Objective To evaluate the role and interaction of IL-1β, TNF-α and TGF-β1 in the pathogenesis of ischemic stroke.Methods The level of IL-1β, TNF-α and TGF-β1 in serum were measured in 46 patients with cerebral infarction in acute phase, 46 patients with carotid atherosclerosisand 22healthycontrols. The level of IL-1β and TNF-α in serum was measured with radioimmunoassay (RIA). The level of TGF-β1 in serum was measured with enzyme linded immunosorbent assay (ELISA). There were serveral statistical methods to be applied. They were analysis of variance, q test, analysis of correlation and t test.Results The level of IL-1β,TNF-α and TGF-β in serum of the patients with carotid atherosclerosis was significantly higher than those of healthy control (P<0.05); The levelofIL-1β, TNF-αand TGF-β1in surmofthepatients with cerebral infraction in acute phase was significantly higher than those of carotid atheroslerosis (P<0.05); The level of TNF-α was positive related with TGF-β1 in surm of the patients with carotid atheroslerosis by correlation analysis (r0.4054, P<0.05); The level of TNF-α was positive related with TGF-β1 in serum of the patients with cerebralinfractioninacutephasebycorrelationanalysis(r0.3499,P<0.05).Conclusions The increasedcytokineswere notmerely the response to cerebral ischemic injury. It had abnormally increased in patients with carotid atherosclerosis. It might play some role in the pathogenesis of ischemic stroke, which suggested that appling TGF-β1 and IL-1β, TNF-α antagonist might be a new way for prevention and curing ischemic cerebral injury in the future.
