健康儿童载脂蛋白E基因表达的研究
Apolipoprotein E gene expression in peripheral blood monocyte in health children
【摘要】 目的 探讨健康儿童外周血单核细胞载脂蛋白E基因表达及其与载脂蛋白E基因型之间的关系。方法 采用竞争性逆转录 -聚合酶链反应方法分析 46例正常健康儿童外周血单核细胞的载脂蛋白E基因表达 ,采用改良的聚合酶链反应 -限制性片段长度多态性方法检测载脂蛋白E基因型。结果 载脂蛋白E基因能在健康儿童外周血单核细胞表达 ,健康儿童载脂蛋白E基因表达量为 (0 47± 0 15 )moles/moleGAPDHmRNA ,载脂蛋白E基因表达量没有明显的性别差异 (P >0 0 5 )。健康儿童载脂蛋白E基因型分布不均 ,E 3 /3 (3 2例 ) ,E 2 /3 (8例 )及E4/3(6例 ) ,未见E 2 /2、E 2 /4、E 4/4。载脂蛋白ε 2、ε 3、ε 4等位基因的频率分别是 0 0 9、 0 85、 0 0 6。与ε 3等位基因携带者比较 ,ε 2等位基因携带者载脂蛋白E基因表达量上调 ,ε 4等位基因携带者的载脂蛋白E基因表达量下调〔分别为 (0 47± 0 12 )moles/moleGAPDHmRNA、 (0 42± 0 10 )moles/moleGAPDHmRNA、 (0 2 8± 0 0 9)moles/moleGAPDHmRNA ,P均 <0 0 5〕。结论 载脂蛋白E基因能在健康儿童周围血单核细胞表达 ,载脂蛋白E基因多态性影响健康儿童外周血单核细胞载脂蛋白E基因表达水平。
【Abstract】 Objective To study the expression status of apolipoprotein(apo)E gene in peripheral blood monocyte in health children and its association with apoE genotypes Methods In 46 health children,apoE gene expression were determined by competi tive reverse transcription-polymerase chain reaction in peripheral blood monocyte ApoE genotypes were determined by means of polymerase chain reaction-restricted fragments length polymorphism Results Expression of apolipoprotein E gene was detected in peripheral blood monocyte in health children The mean of quantity of apolipoprotein E mRNA was(0 37±0 15)moles/mole GAPDH mRNA in health children There were no difference in the apoE gene expression between sexes( P >0 05) The apoE genotypes were unevenly distributed among health children ApoE genotypes frequencies in health children were E 3/3(32/46),E 2/3(8/46), E 4/3(6/46) There were no observation of E 2/2、E 2/4 and E 4/4 ApoE allele frequencies in health children were ε 2=0 09, ε 3=0 85, ε 4=0 06。Compared with apo ε 3 allele,the children with apo ε 2 allele had higher level of apoE gene expression,the children with apo ε 4 allele had lower level of apoE gene expression〔ε 2(0 47±0 12 moles/mole GAPDH mRNA)、ε 3(0 42±0 10 moles/mole GAPDH mRNA)、ε 4(0 28±0 09 moles/mole GAPDH mRNA),respectivly, P <0 05〕 Conclusion Expression status of apoE gene was detected in peripheral blood monocyte in health children ApoE gene polymorphisms could affect the level of apoE gene expression in health children
