二氟甲基鸟氨酸对人肺癌细胞L78细胞生长抑制、凋亡诱导及相关基因的表达调控
Growth inhibition, apoptosis induction and expression regulation of associated genes of the tumor cells L78, treated with DFMO
【摘要】 目的:研究二氟甲基鸟氨酸(DFMO)对人肺癌细胞L78生长及相关基因表达的影响。方法:应用细胞生长曲线、端粒酶活性、细胞形态及DNA电泳观察细胞生长速率及细胞凋亡情况,同时应用Northern blot及免疫组化技术检测相关基因的表达。结果:(1)DFMO处理的L78细胞与对照组相比生长受到抑制,同时,诱导细胞凋亡增多。(2)在DFMO作用下,L78细胞df4基因被诱导表达,同时伴有P21ras表达下调,Fas基因表达的上调。结论:DFMO抑制L78细胞生长,诱导凋亡的作用与其对df4基因的诱导密切相关,DFMO通过对df4的诱导,调控P 21ras和Fas的表达。
【Abstract】 To study the effect of difluromethyl ornithine ( DFMO) on cell growth and expression of the asso-ciated genes of human lung carcinoma L 78. Methods:The cell growth curves, teleomerase activity, morphological ob-servation and DNA electrophoresis were used for assess of the cell growth rate and cell apoptosis, respectively. The gene expression was detected with Northern Blot and immunocytochemical technique. Results:1. The growth of L 78 cells treated with DFMO were inhibited, while the number of the cell apoptosis increased in comparison with control cells. 2. The expression of of 4 in DFMO-treated L 78 cells was induced, in that time, the down-regulated expression of P 21ras genes and the up-regulated expression of Fas gene were detected. Conclusion: DFMO could inhibit the growth and induce the apoptosis of L 78 cells. It would be associated with the induced expression of of 4 gene, by which the expression of P 21ras and Fas gene was regulated.
