问号钩体鞭毛蛋白相关基因分析
Analysis of the flagella protein associated genes in Leptospira interrogens
【摘要】 目的 分析问号钩体黄疸出血群赖型赖株鞭毛蛋白相关基因 ,为进一步实验研究提供信息和线索。方法 使用在线数据库和分析软件NCBI/Blast ,ProDom/Blast等对问号钩体全基因组ORF基因进行诠释 ,对编码蛋白进行在线分析 ,并将问号钩体的鞭毛蛋白基因与E .coliK - 12、伯氏疏螺旋体 (BBU)和苍白密螺旋体 (TPA)的鞭毛蛋白基因进行了比较。结果 确定了问号钩体黄疸出血群赖型赖株染色体上约 5 5个与鞭毛相关蛋白的基因 ,同其他 3个细菌的鞭毛相关基因进行比较分析 ,推测出了钩体鞭毛结构模型图及其装配过程。结论 通过比较 ,不同螺旋体的内生鞭毛形成机制存在差别 ,但是FlgF蛋白缺失可能是其一个共同机制 ;尽管为内生鞭毛 ,问号钩体鞭毛在结构组成和鞭毛装配过程上却与E .coli相似 ;问号钩体的钩形成调控和鞭毛丝结构与E .coli不同 ;在鞭毛形成过程中的转录调控机制也存在差异。
【Abstract】 To annotate the fegella protein associated genes in Leptospira interrogens and to analyze the characteristics of the flagella protein encoded by these genes in order to provide the useful information for further experimental studies, on line database and on line tools, such as NCBI/Blst, and Prodom/Blast were used for the analysis of these genes, and the results were compared with those of Escherchia coli K 12, Treponema pallida and Borrelia burgdoeferi. The experimental results showed that 55 flagella protein associated genes had been recognized in the chromosomes of Leptospira interrogens, serovar icterohaemorrhagiae, serotype Lai. In comaprison with those of other 3 bacterial species, the presumed structural model of flagella proteins in L.interrogens and their assembly processes were established.It was found that the deletion of FlgF was common in the 3 types of spirochaetes, that might account for the production of the periplasmic flagella, and L.interrogens had the similar flagellar structures and assembly processes as that of E.coli,but they were different in filament structure, hook formation and transcription regulation.
