甘露糖在介导二相性白念珠菌粘附中的重要作用
The important role of mannose in mediating adherence of dimorphic Candida albicans
【摘要】 目的研究甘露糖介导的二相性白念珠菌对人口腔颊粘膜细胞的粘附作用。方法分别将孢子相、菌丝相白念珠菌与人口腔颊粘膜细胞进行粘附试验,并用刀豆蛋白A(ConA)作为甘露糖的特异性结合剂,预处理二相性白念珠菌后再进行粘附试验,比较二相性白念珠菌的粘附率;通过气相色谱法测定二相性白念珠菌细胞壁的甘露糖含量,比较二相性白念珠菌细胞壁的甘露糖含量。结果菌丝相白念珠菌对口腔颊粘膜细胞的粘附率显著高于孢子相;用ConA分别预处理菌丝相及孢子相白念珠菌后,再重复进行粘附试验,分别与前述未经ConA处理的粘附率比较,粘附率显著降低。气相色谱法测定孢子相、菌丝相白念珠菌(106cells)细胞壁甘露糖的含量分别为33±2.11μg、151±9.96μg。结论白念珠菌细胞壁上的甘露糖是介导二相性白念珠菌与人口腔颊粘膜细胞粘附的重要物质,且菌丝相白念珠菌细胞壁上的甘露糖含量明显高于孢子相,菌丝相白念珠菌对人口腔颊粘膜细胞的粘附作用强于孢子相。
【Abstract】 To define the important role of mannose in mediating adherence of dimorphic Candida albicans to human buccal epithelial cells (BEC).Yeast forms and hyphal forms of Candida albicans were used respectively to bind with BEC in vitro. Concanavalin A(ConA), a lectin which could recognize mannose, was used to pretreat dimorphic Candida albicans, and then repeated adherence test, the adherence rates of hyphal forms with that of yeast forms was compared. Through Gas chromatographic(GC) assay of mannose in cell walls of dimorphic Candida albicans, the mannose mediated adherence of dimorphic Candida albicans to BEC was studied.The results showed that adherence rates of hyphal forms were higher than that of yeast forms. After treating the hyphal forms and yeast forms with ConA and repeating the adherence test, the adherence rates reduced remarkably. Compared with the adherence rates of ConA treated,and that without con A, the difference is remarkable. GC assay of mannose in cell walls of yeast form and hyphal forms of Candida albicans(10 6 cells) were 33±2.11μg, 151±9.96μg, respectively. It coududes that mannose in cell walls of Candida albicans is important to mediate the adherence of dimorphic Candida albicans to BEC and the content of mannose in cell walls of hyphal forms was much higher than that of yeast forms. The adherence of hyphal forms of Candida albicans to BEC was stronger than that of yeast forms.
