HCMV感染对人神经胶质瘤细胞HOXB1~4及HOXB9基因表达影响的研究
Influence of human cytomegalovirus infection on the expressions of HOXB1-4 and HOXB-9 genes in human glioma cells
【摘要】 目的 研究人神经胶质瘤U2 5 1细胞株的HOXB基因表达状态及人类巨细胞病毒 (HCMV)感染对U2 5 1细胞HOXB1,HOXB2 ,HOXB3,HOXB4及HOXB9基因表达的影响 ,以探讨HCMV致畸的可能机制。方法 应用半定量逆转录 -聚合酶链反应 (RT -PCR)技术检测U2 5 1细胞感染HCMV和 /或全反式维甲酸 (ATRA)处理前后HOXB1,HOXB2 ,HOXB3,HOXB4和HOXB9基因的相对表达水平。结果 U2 5 1细胞表达HOXB1,HOXB2 ,HOXB3,HOXB4及HOXB9基因 ;HCMV感染后 ,U2 5 1细胞HOXB1基因表达明显上调 ,HOXB2和HOXB4基因表达轻微上调 ,HOXB3基因于HCMV感染后表达被抑制 ,HOXB9基因的表达在HCMV感染后不同时间无明显变化 ;ATRA能显著上调HCMV感染后U2 5 1细胞HOXB1基因的表达。结论 HCMV感染能诱导U2 5 1细胞HOXB基因表达改变 ,HCMV有可能通过调控HOXB基因表达异常引起胚胎发育畸形。
【Abstract】 To investigate the expressions of HOXB-1-4 and HOXB-9 genes in human glioma cell strain U251,and the influence of human cytomegalovirus (HCMV) infection on the expressions of these genes in order to explore the possible mechanism of HCMV-induced malformations,the expressions of HOXB genes in U251 cells infected with HCMV and/or treated with ATRA were determined by semi-quantitative RT-PCR.It was found that the expression of the HOXB-1 gene was significantly increased,and those of HOXB-2 and HOXB-4 genes were slightly up-regulated after HCMV infection,but that of the HOXB-3 gene was significantly inhibited.The expression of HOXB-9 gene showed no significant changes.When treated with ATRA,the expression of HOXB-1 in U251 cells infected with HCMV was significantly increased.It concludes that the abnormal expressions of the HOXB genes induced by HCMV may play an important role in HCMV-induced abnormal embryogenesis.
