异种移植胎肠细胞介导排斥反应的免疫学机制
Immunological mechanism of the cellular mediated xenograft rejection in the rat free fetal bowel to mouse transplantation
【摘要】 目的 :用大鼠游离胎肠移植于小鼠模型研究细胞介导排斥反应 (CMXR)的免疫学机理。方法 :观察同种和异种胎肠游离移植各组移植物的病理改变和浸润细胞的表型 ,并用原位杂交方法检测移植物内Th细胞因子mRNA表达。结果 :( 1)移植胎肠的存活率同种移植组高于异种移植组 (P <0 .0 1) ;( 2 )异种移植胎肠主要发生细胞介导的排斥反应 ;( 3)异种移植物内浸润细胞主要为CD4 +T细胞和ED2 +MΦ ,其IL - 4、IL - 5和IL - 10mRNA阳性细胞均明显高于IFN -γmRNA阳性细胞 (P<0 .0 1)。结论 :CMXR与同种移植急性排斥反应存在本质差异 ,其特点是排斥程度强烈 ,异种移植胎肠内以CD4 +T细胞和ED2 +MΦ浸润为主 ,且Th2细胞功能增强。大鼠至小鼠游离胎肠移植模型是研究CMXR的较为理想的动物模型
【Abstract】 Objective:To observe immunological mechanism of CMXR in the rat free fetal bowel transplant to mouse.Methods:Two groups were established including allogroup and xenogroup.Pathological changes of grafts in each group were examined.Expression of TH type cytokines mRNA and phenotypes of infiltrating cells intragraft were detected by in situ hybridization histochemistry and immunohistochemistry methods respectively.Results:(1)Survival rates of allogroup transplants were higher than those of xenogroup( P <0.01).(2)Rejection of xenografts in this model is CMXR.(3)The main infiltrating cells were CD4 +T cells and ED2 +MΦ in xenografts.Expression of mRNA for Th2 type cytokines:IL-4,IL-5,and IL-10 was higher than that for Th1 type cytokine IFN-γ in xenografts.( P <0.01)Conclusion:These results suggest that:(1)CMXR is much stronger than allograft rejection,in which CD4 +T cells and ED2 +MΦ are the major infiltrating cells and the Th2 type cytokines are higher in xenografts that those of in allografts.(2)The model of rat free fetal bowel to mouse transplantation is able to study on CMXR and is easy to operate.
