西乐葆预处理对大鼠脑缺血再灌注损伤ICAM-1表达及白细胞浸润的影响

【摘要】 目的:研究大鼠局灶性脑缺血再灌注损伤的炎症反应机制以及新型抗炎镇痛药物西乐葆(塞来希布)对炎症反应的影响。方法:雄性SD大鼠随机分成实验组(西乐葆灌胃)、实验对照组(安慰剂灌胃)和假手术组,用Longa-Zea氏线栓法复制脑缺血再灌注模型。用2%红四氮唑(TTC)染色测量梗死体积;免疫组织化学法和HE染色观察再灌注不同时间细胞间粘附分子(ICAM-1)阳性表达及浸润白细胞计数。分别比较实验组和实验对照组不同再灌注时间点ICAM-1表达和浸润白细胞计数:比较相同再灌注时间点实验组和实验对照组ICAM-1表达和浸润白细胞计数。结果:(1)实验组梗死体积明显小于实验对照组,(2)脑缺血再灌注后,脑缺血坏死区周边微血管内皮细胞ICAM-1表达及白细胞浸润增多,并于24小时达高峰;相同再灌注时间点实验组ICAM-1表达及白细胞浸润明显低于实验对照组。(3)ICAM-1表达与白细胞浸润及梗死体积的变化呈现同步性。结论:急性炎症反应与局灶性脑缺血再灌注组织损伤有关,ICAM-1介导白细胞和内皮细胞粘附及向脑缺血区浸润过程,参与了脑缺血再灌注损害过程。西乐葆可抑制局灶性脑缺血再灌注时的炎症反应,具有脑保护作用。

【Abstract】 Objective: To study reaction mechanism of inflammation in reperfusion injury after focal cerebral infarction of rat and the effect of Celebrex on inflammation reaction. Methods: Male SD rate are randomly divided into experiment group (feeding Celebrex via nose), contrast group (feeding comforter via nose), pseudo- operation group. To duplicate reperfusion model after cerebral ischemia with Longa- zea’s string embolization method, take picture after stain with 2% TTC, measure volume of infarction area with picture analysis system, observe positive express of ICAM- 1 in different reperfusion time point and leucocytic infiltrate counting with immunochemistry and HE staining method, respectively compare express of ICAM- 1 and leucocytic infiltrate counting in different reperfusion time point between experiment group and contrast group, compare express of ICAM- 1 and leucocytic infiltrate counting in the same reperfusion time point between experiment group and contrast group. Results: (1) Infarction volume of experiment group is apparently less than that of contrast group. (2) After reperfusion, express of ICAM- 1 in microvessel endothelial cell around necrosis area secondary to cerebral ischemia and leucocytic infiltrate increase, achieve summit in 24 hours, in the same reperfusion time point, express of ICAM- 1 and leucocytic infiltrate in experiment group are apparently lower than that of contrast group, express of ICAM- 1 and leucocytic infiltrate summit are postponed. (3) Express of ICAM- 1 has obviously correlation with leucocyte infiltration. Conclusions: ICAM- 1 mediate adhesion of leucocyte and endothelial cell and mediate process of infiltrating cerebral ischemia area of leucocyte. ICAM- 1 and leucocytic infiltrate participate process of reperfusion injury after cerebral ischemia. Acute inflammation reaction has relation with reperfusion injury after focal ischemia. Celebrex is of effect of inhibiting inflammation reaction in. reperfusion after focal cerebral ischemia.
