趋化因子受体CXCR4在人肺癌高转移细胞株的表达和意义
Expression and function of CXCR4 in 95D,a human lung cancer cell line with high metastatic potential
【摘要】 目的 :以人肺癌高、低转移细胞株 95D、95C为研究对象 ,研究趋化因子受体CXCR4的表达及其在肿瘤细胞体外转移潜能中的作用和意义。方法 :采用RT PCR检测 95D、95C细胞CXCR4mRNA的表达情况 ;以PMA活化肿瘤细胞 ,研究CXCR4mRNA表达水平与细胞活性状态的关系 ;应用钙离子内流实验验证其表达是否具有功能 ;通过趋化实验观察CXCR4特异性配件SDF 1α和裸鼠组织匀浆液对 95D细胞的趋化迁移作用 ;通过MTT法测定 95D细胞对SDF 1α作用的增殖反应。结果 :95D细胞功能性地高表达趋化因子受体CXCR4 ,且其表达水平与细胞活性状态有关 ;CXCR4特异性配件SDF 1α和裸鼠肺、淋巴结组织匀浆均可在体外趋化 95D细胞的迁移 ,SDF 1α还可促进 95D细胞的增殖。结论 :95D细胞功能性高表达趋化因子受体CXCR4可能与人肺癌细胞株 95D的体外高转移潜能有关
【Abstract】 Objective:To study the expression and function of chemokine receptor CXCR4 in human lung cancer cell line 95D and 95C with high and low metastatic potential.Methods:Chemokine receptor CXCR4 mRNA was detected by RT-PCR,and the relationship between CXCR4 and 95D cells activation was investigated by PMA;Ca 2+ influx assay was performed to determine the functionally expression of CXCR4 in 95D cells;migration response to CXCR4 particular ligand-SDF-1α and organ-derived protein of nude mice were detected by chemotaxis assay;effect of SDF-1α on 95D cells proliferation was detected with MTT method. Results:Chemokine receptor CXCR4 was functionally and highly expressed on 95D cells,and the expression level of CXCR4 mRNA was vaired when 95D cells were in resting,moderate activation or fully activation conditions.The specific ligand of CXCR4-SDF-1α,also organ-derived protein form nude mice lung and lymph nodes,induced migratory response of 95D cells.Moreover,SDF-1α could promote proliferation of 95D cells.Conclusion:CXCR4 was functionally and highly expressed on 95D cells and the interaction of its ligand SDF-1α with CXCR4 was possibly assiociated with tumor metastasis potential in vitro.
