CⅡ诱导RA患者关节滑液中自身反应性T细胞的免疫学特性分析

【摘要】 目的 :探讨CⅡ诱导的RA患者关节滑液中自身反应性T细胞免疫学特性。方法 :分离RA患者关节滑液中SFMC ,用CⅡ刺激后分析反应性的T细胞频率 ;用ELISA方法检测CⅡ刺激SFMC后上清中的IFN γ和IL 4的含量及通过流式细胞检测SFMC表面标志的表达 ;应用半定量PCR分析外周血及滑膜液中TCRVβ基因表达格局。 结果 :SFMC中CⅡ反应性的T细胞频率高于PBL ;IFN γ的含量在CⅡ刺激SFMC后上清中较高 ,而在未刺激中较低 (P <0 0 1) ,在PBL中更低 (P <0 0 5 ) ;SFMC上CD8及CXCR3表达多于PBMC(P <0 0 5 ) ;外周血T细胞表达绝大多数Vβ基因 ,而滑膜液中TCRVβ基因取用呈现明显的不均衡性。结论 :CⅡ反应性T细胞在SFMC中占优势 ,可能是介导RA发病的主要原因

【Abstract】 Objective:To study the immunological characterization of type II collagen (CⅡ) auto-reactive T cell in patients with RA.Methods: PBL and synovial lymphocyte infiltrates were isolated from peripheral blood and synovial fluid (SF) sample. The frequency of auto-reactive T cells stimulated by CⅡ was analyzed. Interferon-γ (IFN-γ) and interleukine-4 (IL-4) were measured by enzyme-linked immunsorbent assay in the SF, sera and culture supernatants of synovial fluid mononuclear cells (SFMC) that had been stimulated with CⅡ. The phenotype of cells were determined by flow cytometric analysis. TCR Vβ gene expression in peripheral blood and SF of RA were analyzed by semi-quantitatively PCR.Results:The frequency of T cell responses to CⅡ in SFMC was higher than that in PBMC ( P <0 05). The levels of IFN-γ was higher in culture supernatants of SFMC than that in SF (P <0 05). And it was higher in SF than that in sera (P <0 05). CD8/CXCR3 expression on SFMC was higher than that on PBMC ( P <0 05). The usage of most TCR Vβ gene subfamilies was found in peripheral blood and several TCR Vβ gene subfamilies was selectively used in SF of RA. Conclusion:CⅡ reactive T cell was more prevalent in SFMC than that in PBMC from RA. The usage of TCR Vβ gene subfamilies expression in SF of RA may be drived by CⅡ, which play an important role perpetuation in the inflammatory process in RA.
