大鼠肝缺血再灌注核因子-κB活性变化的实验研究

【摘要】 目的 研究肝缺血再灌注(I/R)时核转录因子 κB(NF κB)的活性变化及作用.方法 采用肝I/R模型,用RT PCR技术和免疫组化法分别检测肝I/R后不同时期NF kB亚单位及其抑制因子κBα(I κBα)的mRNA和蛋白质的表达.结果 大鼠肝I/R后1hp65蛋白质活性增高,4h达到高峰,32h仍维持较高水平(P<0.05).而NF κBp65mRNA的表达各组间均无明显改变(P>0.05);I κBα蛋白量于再灌注1h即下降,2h达到最低(P<0.05),以后逐渐恢复正常,I κBαmRNA的表达与其蛋白质的表达相一致(P<0.05).结论 大鼠肝I/R早期肝组织细胞中存在NF κBp65的激活及其I κBα的减少,NF κB的激活可能参与了肝I/R的损伤过程.

【Abstract】 ObjectiveTo observe the activity of nuclear factor-κB during hepatic ischemia reperfusion(I/R) and evaluate its effect.MethodsThe model of hepatic I/R was established and the immunohistochemical technique was used to detect the activity of NF-κB p65 and inhibitory factor-κBα(I-κBα) during different periods of I/R. Their mRNA expressions were also measured with RT-PCR.ResultsNF-κB p65 activity significantly increased after the reperfusion, peaking at 4h, and remained higher level within 32h, but the mRNA expression didn’t change much; I-κBα and its mRNA expression began to decrease at 1h after the reperfusion, peaking at 2d after the reperfusion ( P <(0.05)), and then gradually restored.ConclusionBoth activated NF-κB and decreased I-κB exist in the hepatic tissues during I/R, the activated NF-κB may be involved in the I/R injury.
