大豆秸秆酶水解的影响因素的研究
The Affecting Factors for Enzymatic Hydrolysis of Soybean Straw
【摘要】 为了从大豆秸秆中提取生物降解性塑料的原料 -乳酸 ,对大豆秸秆纤维素酶水解条件进行了研究。酶水解的影响因素主要为秸秆的预处理条件 ,酶水解pH值、反应温度、反应时间、底物浓度、酶用量。研究结果表明 ,较适宜的预处理条件为大豆秸秆粉碎至 14 0目 ,10 %氨水处理 2 4h。经过预处理后秸秆酶水解最佳工艺条件为 :pH =4 .8,温度为 4 5℃ ,反应时间为 2 8h ,底物浓度为 5 % ,酶用量为 4 5 0FPU/g(秸秆 ) ,大豆秸秆酶水解率为 2 8.6 3%。研究结果为大豆秸秆酶解液乳酸发酵实验提供了理论依据
【Abstract】 For making lactic acid as a material to produce biodegradable plastic from soybean straw, the affecting factors during the enzymatic hydrolysis of soybean straw were studied.The major affecting factors are the pretreatment conditions,including pH value, reaction temperature,time,substrate concentration and enzyme dosage. It shows that the combination of straw grinding and ammonia pretreatment greatly influences the enzymatic hydrolysis of soybean straw to produce sugar. The optimum pretreatment conditions were obtained as follows: grinding soybean straw to 140mu, and treating with 10% ammonia solution for 24h. The experiment results show the following optimum conditions for the enzymatic hydrolysis: pH 4.8, temperature 45℃, hydrolysis time 28h, substrate concentration 5%, and enzyme dosage 450FPU/g(soybean straw).
