SD大鼠肌腱冷冻维持温度选择的实验研究

【摘要】 目的 :探讨不同维持温度冷冻预处理超深低温保存对肌腱超微结构的影响 ,为同种异体肌腱保存移植提供依据。方法 :实验组设计维持温度为 -4 5℃、-5 0℃、-5 5℃、-60℃、-65℃、和 -70℃组 ,以 15 %二甲基亚砜为低温保护剂 ,对SD大鼠肌腱进行上述温度冷冻预处理 ,液氮中保存 1周 ,HE染色组织学观察和透射电镜肌腱超微结构观察。结果 :电镜下 -4 5℃组胶原纤维排列较稀疏 ;-60℃组多数腱细胞胞浆水肿 ,核膜不完整 ;-65℃组胞浆内细胞器破坏 ;-70℃组部分腱细胞坏死脱落。 -5 0℃组和-5 5℃组与对照组之间差异不明显 ,肌腱组织超微结构得到良好保持。结论 :肌腱组织冷冻预处理的维持温度在 -5 0℃~ -5 5℃之间 ,-5 0℃是肌腱组织冷冻预处理的最佳维持温度值 ,可获得具有大部分生物活性的肌腱。

【Abstract】 Objective: To investigate the effect of preliminary treatment at a variety of temperature on the ultrastructure of frozon tendon preserved at ultra-low temperature and provide the basis for allograft tendon transplantation. Methods: The tendons taken from SD rats were treated at -45℃?-50℃? -55℃?-55℃?-60℃?-70℃with 15% DMSO taken as low temperature protective reagent, and then preserved in liquid nitrogen for one week. The frozen tendon was sliced and dyed by HE and studied under light microscope and TEM (transmission electron microscopy). Results: The Arrangement of collagen fibers in tendon at -45℃ under TEM was sparse and scattered. At -60℃, most of tendons showed cytoplasm edema and the karyotheca was not complete. At -65℃, the cellular organelle was destroyed. At -70℃, some tendinous cell died . There was no significant difference between the groups of -50℃?-55℃ and control group, the ultrastructures of the frozen tendon were well preserved. Conclusions: he maintained temperature is -50℃ to -55℃ for the preliminary treatment of the frozen tendon. -50℃ is the optimal maintained temperature which can ensure the biological activity of tendon as far as possible.
