猪源致病性沙门氏菌四环素耐药基因tetC的PCR扩增及序列分析
PCR amplification and nucleotide sequences analysis of tetC gene of pathogenic Salmonella from swine
【摘要】 本研究对 16株猪致病性沙门氏菌的四环素耐药性进行了检测 ,结果表明 ,沙门氏菌对四环素类抗生素产生了广泛的耐药性 ,耐药率达 10 0 %。对四环素耐药基因 tet C进行 PCR扩增 ,结果在 12株沙门氏菌的质粒上扩增出特异性产物 ,与药敏试验结果阳性符合率 75 % ,具有较高的检出率 ;序列分析结果表明 tet C的核苷酸同源率较高 ,达 97.7%~ 98.7% ,建立 tet C基因的 PCR检测技术 ,为沙门氏菌对四环素耐药性的分子流行病学监测提供了有效的途径。
【Abstract】 The detection of tetracyclines-resistance of 16 Salmonella isolates showed the resistance rate was 100%. The tetC gene of 16 Salmonella strains was amplified by polymerase chain reaction. Twelve strains with this gene were located on plasmids. Compared to the results of the drug-susceptibility tests, the method of PCR showed 75% concordance in positive rate. The method of using PCR to detect tetC gene was established, and it could be applied to the molecular epidemiology surveillance of tetracyclines-resistance.
