PCI-hTGF-β1转染体外培养成人退变椎间盘细胞及其表达产物的测定

【摘要】 目的:测定真核表达载体PCI-hTGF-茁1转染体外培养成人退变椎间盘细胞的表达产物hTGF-茁1,为椎间盘退变的基因治疗提供实验基础。方法:用免疫组织化学方法对转染细胞进行表达产物测定;用VIDAS数据分析系统对实验数据进行定量分析。结果:原代纤维环细胞转染中真核表达载体PCI-hTGF-茁1阳性细胞产物的光密度值为PCI组的3.49~3.56倍,为未转染组的3.43~3.55倍;原代髓核细胞转染中真核表达载体PCI-hTGF-茁1阳性细胞产物的光密度值为PCI组的3.46~3.69倍,为未转染组的3.33~3.63倍。结论:真核表达载体PCI-hTGF-茁1可以成功转染体外培养的成人退变椎间盘细胞,并可获得hTGF-茁1基因产物的高表达。

【Abstract】 Objective:To provide experimental foundation for the clinical practice of gene therapy of disc degeneration diseases by determining the expressive product of hTGF-β1.Method:Expressive product of the transfected cell was analyzed with immunohistochemical staining method and with VIDAS Software analysis system,the data was evaluated quantitatively.Result:The OD value of the positive cells production in the eukaryotic vector PCI-hTGF-β1 group was 3.49~3.56 times and 3.43~3.55 times of that in PCI group and untransfective group respectively in the original annulus fibrosus cells transfection.The OD value of the positive cells production in the eukaryotic vector PCI-hTGF-β1 group was 3.46~3.69 times and 3.33~3.63 times of that in PCI group and untransfective group respectively in the original nucleus pulposus cells transfection.Conclusion:Eukaryotic vector PCI-hTGF-β1 constructed in our laboratory can transfect adult degenerative disc cells cultured in vitro.The gene product of hTGF-β1 can be expressed.
