用髓鞘碱性蛋白建立豚鼠变态反应性脑脊髓炎模型
The Establishment of Experimental Allergic Encephalomyelitis Model on Guinea Pig
【摘要】 目的 :用髓鞘碱性蛋白 (MBP)诱导实验性变态反应性脑脊髓炎 (EAE)豚鼠模型。方法 :提取MBP并经SDS 聚丙烯酰胺凝胶电泳 (SDS PAGE)鉴定后 ,用MBP +完全弗氏佐剂 (CFA)免疫豚鼠 ,观察豚鼠的临床症状和中枢神经系统 (CNS)的病理改变。结果 :实验组豚鼠均表现出典型的EAE症状 ,发病率为 10 0 % ,并在光镜下可见CNS血管周围炎性细胞浸润和白质脱髓鞘病理改变。结论 :用MBP +CFA成功地诱导了豚鼠的EAE模型 ,具有模型稳定、发病率高、动物价廉易得的优点 ,是研究多发性硬化 (MS)的理想动物模型
【Abstract】 Objective: To induce the EAE model on Guinea pig wi th Myelin Basic Protein(MBP). Methods: The MBP was extracted and identified by SDS-PAGE,and then the Guinea pig was immunonized with MBP+CFA. B oth the clinical symptoms and histopathologic changes of the central nervous sys tem were observed.Results: The Guinea pigs in the experimental gro up all demonstrated the typical symptoms of EAE with the incidence of 100%.At the same time,the infiltration of mononuclear cells in the perivasculars and the de-myelinationin of the white matter were observed through optical microscope .Conclusion:The EAE model of Guinea pig was induced successfully w ith MBP+CFA; furthermore,the model was stable and had high incidence as well as the Guinea pig so was cheap and can be bought easily,that this mode l might be an ideal zoo-model to study the MS. [
