转反义vp1基因水稻再生苗的获得及分子鉴定

【摘要】 应用超级双元载体和农杆菌介导转基因技术 ,试图将反义vp1基因引入到容易穗萌的保持系冈 46B和容易诱导愈伤组织的籼稻品种R893与容易转化的粳稻品种粳 16中 ,结果冈 46B获得再生苗 41株 ,R893获得再生苗 43株 ,粳 16获得再生苗 2 6株。经PCR检测 ,仅粳 16中有 2株携带潮霉素抗性基因 ,其中 1株携带反义vp1基因 ;冈 46B和R893中未能获得转基因再生苗。

【Abstract】 Anti-sense vp1, a potential preharvest sprouting (PS) inhibitor, was intended to be introduced into Gang 46 B, a PS susceptable B line, R893, an easily callus-inducible indica R line and Jing 16, an easily transformable japonica variety by Agrobacterium-mediated transformation using super binary vector. 41 regenerated seedlings of Gang 46 B, 43 of R893 and 26 of Jing 16 were obtained. Only 2 seedlings in Jing 16 were confirmed carrying hygromycin-resistant gene and one of them carrying anti-sense vp1while no transgenic seedlings were found in Gang 46B and R893, by PCR test using specific primers of hygromycin-resistant gene and anti-sense vp1, respectively.
