核酸疫苗VR1012/Sj31粘膜免疫诱导小鼠抗攻击感染的研究

【摘要】 目的 探讨日本血吸虫核酸疫苗VR10 12 /Sj3 1粘膜免疫诱导小鼠抗攻击感染的保护力。 方法 将实验小鼠随机分为 5组 ,每组 12只。A组为单磷脂 (MPL)组 ,B组为VR10 12组 ,C组VR10 12 +MPL组 ,D组为VR10 12 /Sj3 1组 ,E组为VR10 12 /Sj 3 1+MPL组。滴鼻免疫。在第 3次免疫后 2周 ,每只鼠经腹部感染 40± 1条尾蚴 ,45d后宰杀 ,观察减虫率和减卵率。在初次免疫前和攻击感染前尾静脉采血并收集粪便 ,ELISA检测血清内特异性IgA、IgG及粪内sIgA水平。  结果 与对照组相比 ,VR10 12 /Sj 3 1及VR10 12 /Sj 3 1加MPL滴鼻免疫均可引起小鼠血清IgG、IgA和粪内sIgA升高 ,且VR10 12 /Sj 3 1加佐剂滴鼻免疫组抗体的增幅 >不加佐剂组。VR10 12 /Sj3 1和VR10 12 /Sj3 1加MPL滴鼻免疫诱导小鼠产生的减虫率分别为 2 1.70 %和 2 8.82 % ,减卵率分别为 2 7.98%和 40 .72 %。二者的减虫率差异无显著性 (P >0 .0 5 ) ,但减卵率后者显著高于前者 (P <0 .0 5 )。 结论 VR10 12 /Sj3 1滴鼻免疫可引起小鼠产生全身和粘膜免疫应答及部分抗攻击感染的免疫保护力。

【Abstract】 Objective To explore the immune protection against challenge infection of Schistosoma japonicum induced by mucosal vaccination of VR1012/Sj31 in mice. Methods VR1012/Sj31 were amplified,extracted and purified by routine methods. Mice were divided into 5 groups each with 12 mice. They were immunized intranasally with monophosphoryl lipid A (MPL),VR1012,VR1012+MPL,VR1012/Sj31,VR1012/Sj31+MPL respectively. The mice were challenged with 40±1 S. japonicum cercaria per mouse 2 weeks after third immunization. 45 days later,mice were killed and perfused,and the adult worms and eggs reduction rates were counted,sera and faeces samples were collected before the first immunization and challenge infection. IgA and IgG in sera and sIgA in faeces were detected by ELISA. Results VR1012/Sj31 induced mucosal and systemic immune response and the levels of specific antibody induced by intrnasal vaccinations of VR1012/Sj31+MPL were higher than that by vaccinations of VR1012/Sj31 only. The worm reduction rate and the egg reduction rate were 21.70%,28.82% and 27.98%,40.72% respectively by the intranasal vaccinations of mice with VR1012/Sj31 and VR1012/Sj31+MPL. The difference of worm reduction rate between two groups was not significant ( P >0.05),and the difference of the egg reduction rate between two groups was significant ( P <0.05). Conclusion VR1012/Sj31 induced mucosal and systemic immune response and partial protection against challenge of S. japonicum by intransal vaccination in mice.
