汉坦病毒M基因区核苷酸序列及G1膜蛋白部分氨基酸共享序列分析

【摘要】 目的对青岛地区31株汉坦病毒分离株M区基因序列测定及基因分型研究,为疫苗接种提供依据。方法从64例肾综合征出血热(HFRS)患者发热期血清标本中提取RNA,应用套式逆转录聚合酶链反应扩增汉坦病毒基因组M片段,G1基因区克隆并测定核苷酸序列和部分共享氨基酸序列,且对测定结果应用DNAStar和Prosis软件进行分析。结果64份标本中测出汉滩型(HTN)6份,占9%;汉城型(SEO)25份,占39%;总阳性率48%。SEO型汉坦病毒间核苷酸序列的差异相对较小,其基因的离散度为03%~89%。HTN型汉坦病毒变异率较高,基因的离散度为26%~112%。结论SEO型汉坦病毒基因型相对保守,而HTN型变异率较高;M区段可编码一个具有稳定的保守抗原表位。

【Abstract】 Objective To study on the M genome sequence and genotyping of thiry one Hantavirus Strains which were applied for inactivated vaccine production in China Methods RNA was extracted from sixty four serum samples of clinically diagnosed patients with hemfever and renal syndrome(HFRS).The RT PCR products was cloned into Tvector ,sequenced and analyzed by using DNASTAR and prosis sofrtware Results In the sixty four serum samples,HTN genotype was six strains (9%)and SEO genetype was twenty five strains (39%).The total positive rate was 48% The variation rate of SEO The genetype was HTN genetype were 0 3%~8 9% and 2 6%~11 2% respectivety Conclusion The SEO genetype was relative conservativeness .The variation rate of HTN genetype was relative higher;M genome may encode a stable conservative antigen epitope
