10-羟基喜树碱对HepG2体外生长及其小鼠肾包膜下移植瘤的影响

【摘要】 目的 探讨 10 羟基喜树碱 (HCPT)对人肝癌细胞HepG2体外生长及其小鼠肾包膜下移植瘤的影响。方法 用 10 HCPT处理HepG2细胞 ,四甲基偶氮唑蓝 (MTT )法检测其生长指数 (GI) ,软琼脂克隆形成试验检测其克隆形成率 ;建立HepG2小鼠肾包膜下移植瘤模型 ,每天腹腔注射 10 HCPT ,第 6天处死动物 ,测量瘤块体积。结果  10~ 3 60mg/L浓度 10 HCPT处理组HepG2细胞GI分别为 84.72 %~ 47.65 % ,与对照组比较明显降低 (P <0 .0 5 )。 90~ 3 60mg/L 10 HCPT处理组HepG2软琼脂克隆形成率分别为 3 8.80 %~ 2 5 .2 0 % ,与对照组比较亦明显降低 (P <0 .0 1) ,两者均具有量效依赖关系。 3× 10 -3 ~ 9× 10 -3 mg/3 0 g剂量 10 HCPT处理组小鼠肾包膜移植瘤体积分别为 2 .93~ 1.88mm3 ,与对照组比较明显缩小 (P <0 .0 1)。结论  10 HCPT对HepG2的体外生长及其小鼠肾包膜移植瘤均具有明显抑制作用。

【Abstract】 Objective To investigate the effects of 10-HCPT on proliferation of human liver cancer cell line HepG2,and growth of its xenograft implanted to the subrenal capsule of normal immunocompetent mouse.Methods The human liver cancer cell line HepG2 was cultured,and was treated by different concentrations of 10-HCPT in vitro.The cell growth index (GI) and the clone formation rate of HepG2 were detected by MTT colorimetric assay and soft agar assay,respectively.The HepG2 cells were implanted to the subrenal capsule of KM mice treated by 10-HCPT through abdominal cavity injection everyday and the growth of the xenografts was measured on the sixth day after transplantation.Results The GI of HepG2 cells treated in vitro by 10-HCPT with concentration ranging from 10 ng/L to 360 ng/L was 84.72% to 47.62%,significantly lower than that of control group (P<0.05).The clone formation rates of HepG2 treated by 10-HCPT with concentrations from 90 ng/L to 360 ng/L were 38.80% to 25.20%,very significantly lower than that of control groups (P<0.01).There were dose-dependent relationships among them.The volumes of xenografts treated in vivo by 10-HCPT from 3×10 -3 mg/30 g to 9×10 -3 mg/30 g weight were 2.93~1.88 mm3,smaller than that of control group very significantly (P<0.01).Conclusion 10-HCPT could inhibit the growth and proliferation of HepG2 in vitro,and decrease the volumes of xenografts in vivo effectively.
