应用微阵列初步探讨63例脑胶质瘤的肿瘤相关基因表达谱

【摘要】 目的应用微阵列考察不同类型脑胶质瘤的肿瘤相关基因表达谱信息。方法术中收集不同类型胶质瘤共63例以及5例正常脑组织,提取总RNA,逆转录成32P标记的cDNA探针,与Atlas人肿瘤微阵列杂交,通过放射自显影获得微阵列杂交图,应用AtlasImageTM1.01a分析肿瘤与正常脑组织之间的肿瘤相关基因差异表达。结果高恶性度胶质瘤的差异表达基因数多,而低恶性度胶质瘤的差异表达基因数少。15个在胶质瘤中差异表达频率最高且表达趋势一致的基因中原癌和抑癌作用基因约各占一半。大部分已知与胶质瘤相关基因的表达与已有报道相符。结论胶质瘤是一种多基因病变,差异表达基因数量与肿瘤的恶性度相关,基因之间存在复杂的相互作用关系,胶质瘤的发生可能是促进和抑制肿瘤作用基因失平衡的结果。

【Abstract】 Objective To detect the cancer-related gene expression profiles of different pathological types of gliomas with cDNA array. Methods 63 samples of different pathological types of gliomas and 5 of the normal brain tissues were collected freshly during operation. After extraction of the total RNA the 32P targeted cDNA probes were converted and then hybridized with Atlas Human Cancer Array1.2. The hybridization maps of cDNA arrays were acquired through autoradiography. The discrepancy between the tumors and the normal brain tissues was analyzed with Atlas ImageTM1.01a. Results More differential expression genes were detected in the malignant gliomas than the low grade and benign ones. 15 genes were differentially expressed in most of the gliomas with the same expression trend  among which the number of oncogenes and the anti-oncogenes were half and half. Most of the known glioma-related genes were consistent with the published reports. Conclusion Gliomas are lesions involving multiple genes. The number of the differential expression genes among which some complex interacting relationships are involved is relative to the malignant grade of gliomas. The gliomas may result from the imbalance between the tumor-facilitating genes and the anti-oncogenes.
