17β-雌二醇对人破骨样细胞RANK、CKmRNA表达的作用

【摘要】 目的 观察 17β 雌二醇 (17β E2 )对破骨样细胞破骨细胞分化因子受体 (RANK)和组织蛋白酶K (CK)mRNA表达的影响。方法 人巨细胞瘤组织经过滤、培养、洗涤、胰酶消化后再经孔径 2 5 μm微过滤网过滤 ,得到纯化的破骨样细胞 ,用RT PCR观察 17β E2 对人破骨样细胞CK及RANKmRNA表达的影响。结果  (1)破骨样细胞具有成熟破骨细胞的表型特征 ,如抗酒石酸酸性磷酸酶和酸性磷酸酶染色阳性、具有溶骨作用 ,可表达CK和RANK ;(2 ) 17β E2 对人破骨样细胞RANKmRNA表达无明显影响 ,但下调CKmRNA表达 (P <0 .0 5 )。结论 破骨样细胞是进行破骨细胞研究的较好细胞来源 ;17β E2 可下调破骨样细胞CKmRNA表达。下调CK基因表达是绝经后雌激素补充治疗的药理机制之一。

【Abstract】 Objective To observe the effects of 17β-estradial (17β-E 2) on the expressions of receptor activator of nuclear factor-kappa B (RANK) and cathepsin K (CK) mRNA in osteoclast-like cells (OCLs). Methods The OCLs were isolated from human giant-cell tumor with 0.25% trypsin, 0.2% type Ⅰ collagenase digestion and Φ25 μm mesh. The effects of 17β-E 2 on the expressions of RANK and CK mRNA in OCLs were assayed by RT-PCR. Results (1) The OCLs showed the phenotypic characteristics of matural osteoclasts such as positive tartrate-resistant acid phosphatase staining and positive acid phosphatase staining, osteolysis and expressions of CK and RANK mRNA. (2) There was no significant effect of 17β-E 2 on the expression of RANK mRNA in OCLs, but 17β-E 2 down-regulated the expression of CK mRNA in OCLs (P<0.05). Conclusion The established new technique for purifying OCLs provides enough amount of OCLs for study. 17β-E 2 down-regulates the expression of CK in OCLs with a decrease in the osteolytic function. Down-regulated expression of CK is one of the important mechanisms of estrogen replacement therapy in estrogen deficiency-induced osteoporosis after postmenopause.
