尼尔雌醇对人成骨样MG63细胞护骨素和骨钙素基因表达的影响

【摘要】 目的 研究尼尔雌醇对人成骨样MG63细胞护骨素 (OPG)、骨钙素 (BGP)基因表达的影响。方法 MG63细胞以不同浓度的尼尔雌醇及雌激素受体纯拮抗剂ICI 182 780分别干预 2 4h、48h及 72h ,用半定量RT PCR法检测OPG、BGPmRNA的表达量。结果 尼尔雌醇可上调MG63细胞OPGmRNA的表达 ,并存在剂量依赖和饱和效应 ,其中以 10 -8mol/L的作用最强。而且OPGmRNA表达有时间依赖性 ,以 48h表达量为最高。尼尔雌醇未显示对MG63细胞BGPmRNA表达的影响。结论 尼尔雌醇上调MG63细胞OPGmRNA表达是其作为抗骨质疏松药物的重要机制。

【Abstract】 Objective To examine the effects of nylestriol on osteoprotegrin (OPG) and osteocalcin (BGP) gene expressions in human osteoblast-like MG63 cells. Methods MG63 cells were cultured with different concentrations of nylestriol (10 -10 -10 -6 mol/L) with or without estrogen receptor pure antagonist ICI 182780 for 24, 48 and 72 h respectively, and then the semiquantitive RT-PCR was performed to detect the mRNA expressions of OPG and BGP in MG63 cells. Results OPG mRNA expression regulated by nylestriol showed a dose-dependent and saturation manners at 24 h and reached the maximal effect in 10 -8 mol/L nylestriol group (1.308±0.016) in MG63 cells. OPG mRNA expression up-regulated by 10 -8 mol/L nylestriol could be blocked by ICI 182780 (ICI 10 -7 mol/L+NYL 10 -8 mol, 0.544±0.040; ICI 10 -8 mol/L+NYL 10 -8 mol/L,0.529±0.023). OPG mRNA expression regulated by nylestriol also exhibited a time-dependent manner, with the maximal expression at 48 h. Different concentrations of nylestriol showed no difference in BGP mRNA expression in MG63 cells. Conclusion Up-regulated OPG mRNA expression is a significant mechanism of nylestriol as an anti-osteoporosis drug.
