微粒子载体携反义MCP-1基因的转染抑制腹主动脉瘤生长的实验研究

【摘要】 目的 探讨微粒子携反义单核细胞趋化蛋白 1(MCP 1)基因局部定位转染对大鼠腹主动脉瘤生长的影响及机制。 方法 将大鼠制成腹主动脉瘤模型后 ,分成微粒子 DNA组和空载粒子对照组、生理盐水对照组 ,分别进行腹主动脉局部定位转染 ,2周后观察腹主动脉直径的变化 ,应用聚合酶链反应 (PCR)观察微粒子作为基因转染载体的可行性 ,应用原位杂交、Western蛋白质印迹等方法观察反义基因转染后对内源性MCP 1基因表达的抑制作用。 结果 基因转染后 2周 ,微粒子 DNA组腹主动脉直径为 (1 79± 0 12 )mm ,明显小于 2个对照组 (P <0 0 1) ,PCR扩增见该组动脉组织提取的DNA中有特异性条带出现 ,动脉壁中MCP 1基因的mRNA和蛋白表达水平明显低于对照组 (P <0 0 1)。 结论 微粒子可有效地用作基因转染的载体 ;反义MCP 1基因的转染可通过抑制内源性MCP 1基因的表达和动脉壁巨噬细胞的浸润而达到抑制腹主动脉瘤生长的作用。

【Abstract】 Objective To explore the influence of microparticle-mediated antisense monocyte chemotactic protein-1 (MCP-1) transfection to the development of abdominal aortic aneurysms and its mechanism. Methods Rats were randomly divided into microparticle-DNA group and two control groups after experimental abdominal aortic aneurysm models were created. Genes were transfected into abdominal aorta after elastase perfusion. Two weeks after surgery, diameter of abdominal aorta was measured and aortic tissue was obtained. PCR was used to evaluate the feasibility of microparticle as a carrier of gene transfection, while tissue hybridization and western blotting were applied to detect the inhibitory effect of endogenous MCP-1. Results Two weeks after transfection, the abdominal aortic diameter of microparticles-DNA group was (1.79±0.12)mm, smaller than that of control groups (P < 0.01); and specific band of antisense MCP-1 was shown by PCR. The expression of MCP-1 mRNA and protein in microparticle-DNA group was less compared with that in control group (P<0.01). Conclusions Microparticles can be effectively acted as the carrier of gene transfection. Antisense MCP-1 transfection can inhibit the development of abdominal aortic aneurysms via inhibiting the endogenous MCP-1 and macrophage infiltration.
