木聚糖酶酶活影响因素的研究

【摘要】 研究了两种耐温耐碱的商业木聚糖酶酶活的影响因素:木聚糖酶A和木聚糖酶B酶反应的最适pH值为7.0,酶反应的最适温度分别为65℃和70℃。Fe3+在低浓时对这两种酶有激活作用,Fe3+在高浓时则有抑制作用。Al3+对木聚糖酶A有抑制作用,对木聚糖酶B有激活作用。Cu2+对木聚糖酶A和木聚糖酶B都表现为抑制作用。Mn2+对木聚精酶A有激活作用,对木聚精酶B有抑制作用。

【Abstract】 The influential factors of xylanases (EC3.2.1.8) activity were studied. Optimum pH for both xylanase A and xylanase B wns 7.0,the optimum temperature for xylanase A and xylanase B was 65℃ and 70℃ respectively. The activity of both A and B enzyme wasactivited by Fe3+ at low concentration, but inhibited by Fe3+ at high concentration. Al3+ had inhibition on xylanase A, but a promoting effecton xylanase B. The activity of both xylanase A and xylanase B was inhibited by Cu2+. Mn2+ activated xylanase A, but inhibited xylanase B.Xylanase A had Km of 5.57 mg/mL and Vmax of 1.98×104μmol/ (mL· min ). The Km and Vmax for xylanase B were 6.09 mg/mL and 3.64×104μmol/ (mL·min).
