固相萃取-高效液相色谱法测定肝移植病人血清中麦考酚酸浓度
RP-HPLC determination of mycophenolic acid in serum of liver transplant patients with solid-phase extraction
【摘要】 目的 建立血清中麦考酚酸 (霉酚酸 )的高效液相色谱检测法 ,并监测肝移植病人霉酚酸血药浓度。方法 应用OASIS固相萃取小柱提取血清中霉酚酸 ,采用反相高效液相色谱法二极管阵列检测器检测 ,色谱柱为SymmetryC1 8(2 5 0mm× 4 6mm ,5 μm) ,流动相为乙腈 2 5mmol·L- 1 KH2 PO4 缓冲液 (5 0∶5 0 ) ,pH为 4 5 ,流速为 1 0mL·min- 1 ,柱温 2 5℃ ,检测波长 2 10nm。结果 霉酚酸在 0 1~ 10 0 μg·mL- 1 内定量线性良好 (r=0 9999) ,精密度和回收率均良好 ,成功用于肝移植病人霉酚酸血药浓度测定。结论 本方法适用于肝移植病人霉酚酸血药浓度的测定 ,但临床测定结果较文献报道的肾移植病人口服麦考酚酸酯后MPA血药浓度数据低 ,有待进一步研究。
【Abstract】 OBJECTIVE To establish an analytical method to determine the serum concentrations of mycophenolic acid in patients with liver transplantation. METHODS The mycophenolic acid in serum was extracted by Waters OASIS solid-phase extractor. The analytical column was Symmetry C18 (250 mm×4.6 mm, 5 μm) and column temperature was 25℃. The mobile phase was acetonitrile-2.5 mmol·L-1 KH2PO4 solution (pH=4.5) (50∶50) with flow rate of 1.0 mL·min-1. The detection wavelength was 210 nm. RESULTS The method was linear within the range of 0.1~10.0 μg·mL-1 (r=0.999*!9), and the precision and recoveries were good. It had been used successfully to determine clinical samples. CONCLUSION This method could be used to determine mycophenolic acid in serum of patients with liver transplantation. But the concentrations of liver transplantation patients were lower than those of renal transplant patients.
