高压氧对aGVHD小鼠皮肤、肺、肠组织中T淋巴细胞及其亚群浸润和黏附分子表达的作用

【摘要】 目的 :观察高压氧对骨髓移植时急性移植物抗宿主病 (aGVHD)的作用机制。方法 :应用直线加速器排空C57BL/6小鼠骨髓后 ,注入BALB/C小鼠骨髓与脾淋巴细胞的混合液 ,复制aGVHD模型。应用免疫荧光电镜检测经高压氧、环孢素A(CsA)、氨甲喋呤 (MTX)处理后 ,小鼠皮肤、肺、肠组织T淋巴细胞 (CD3+ 、CD4 + 、CD8+ 细胞 )及表达黏附分子CD11a、CD18的细胞浸润情况。结果 :高压氧组小鼠 11d的生存率 (9/10 )高于异基因骨髓移植组 (8/19) ;高压氧组小鼠T细胞及其亚群的数量降低 ,黏附分子CD11a、CD18的表达降低。结论 :高压氧可能通过降低T细胞及其亚群的数量并降低黏附分子CD11a、CD18的表达 ,而发挥其明显的抗aGVHD作用

【Abstract】 Objective: To observe the effects of hyperba ric oxygen(HBO) on T lymphocyte subsets and adhesion molecule in skin, lung, and intestine in mice with acute graft-versus-host -disease(aGVHD), and to study the mechanism of HBO antagonizing aGVHD.Methods:The C 57 BL/6 mice were exposed to radiation with linear accelerator, then given the si ngle cell suspensions which were obtained from bone marrow and spleens of the B ALB/C mice. The model of aGVHD were duplicated. The receptors were treated by us ing HBO and CsA plus MTX. The condition of T lymphocyte subsets and adhesion mol ecule were determined by using immumofluorescence electron microscope. Results:The survival rate of HBO group (9/10)was much higher than that of allo-BMT group(8/19). The quantity of T cell and the subsets in HBO group decreased. The expression of adhesion molecule decreased too. Conclusio n:HBO could inhibit aGVHD significantly. This effect of HBO may be r elated to the decreased quantity of T cell and its subsets and decreased express ion of adhesion molecule.
