OK-432对5-氟尿嘧啶在小鼠体内药代动力学的影响

【摘要】 目的 :通过检测 5 氟尿嘧啶 (5 FU)在小鼠体内的血药浓度 ,分析OK 4 32对 5 FU在小鼠体内药代动力学参数的影响。方法 :小鼠腹腔注射后定时采血 ,研究单用 5 FU及其联用OK 4 32后的药代动力学 ,并用反相高效液相色谱法精密检测血浆中 5 FU的浓度 ,用“3p97”程序计算药动学参数。 结果 :联用OK 4 32后 ,5 FU的时间浓度曲线下面积AUC增大 ,进入血循环总药量增加 ,消除速率半衰期t1 /2 β增加 ,作用时间延长。结论 :联用OK 4 32可使 5 FU在体内吸收减慢 ,达峰时间延迟 ,但吸收总量增加 ,消除延缓。两药联用时注意调整剂量和给药间隔

【Abstract】 Objective: To study the effect of OK 432 on pharmacokinetics of 5 fluorouracil(5 FU). Methods: The concentration of 5 FU in plasma of mice was detected with reverse phase high performance liquid chromatography(RP HPLC)and the data were dealt with “3p97” program using computer. Results: The area below the curve of 5 FU was increased when OK 432 was used in combination. The total amount of drug in circulation increased. The half life of elimination was increased and the action time was prolonged. Conclusion: Compared with 5 FU used alone, its absorbance phase time was increased when OK 432 used in combination. The top concentration was decreased and the area under the curve was increased, but elimination process was prolonged. The dosage and interval of administration should be paid more attention.
