核因子κB与细胞周期蛋白D1在骨肉瘤中的表达及相关性研究

【摘要】 目的 :探讨骨肉瘤细胞中核因子κB(NF κB)及细胞周期蛋白D1(cyclinD1)的表达及其意义。 方法 :应用免疫组织化学SABC法 ,检测 4 6例骨肉瘤、15例骨软骨瘤和 5例正常骨组织中NF κBp6 5亚单位和cyclinD1表达 ,并比较NF κB表达在骨肉瘤组织学分级、分型间的差异及其与cyclinD1表达的关系。 结果 :NF κBp6 5亚单位显著表达于骨肉瘤细胞中 (6 7.4 % ) ,而在正常骨组织和骨软骨瘤中未见表达 ;NF κBp6 5表达与骨肉瘤组织学分级无关 ,在骨内高分化型组表达显著高于普通中心型组 (P<0 .0 5 )。cyclinD1在骨肉瘤中表达率为 73.9% ,显著高于骨软骨瘤组 (33.3% ,P <0 .0 5 ) ;骨肉瘤中NF κB p6 5与cyclinD1蛋白表达显著正相关 (r =0 .4 6 3,P <0 .0 1)。结论 :NF κBp6 5在骨肉瘤细胞中异常活化 ,并与cyclinD1过表达密切相关 ,其可能通过上调cyclinD1表达参与骨肉瘤的发生发展。

【Abstract】 Objective: To study the expressions and significance of nuclear factor-kappa B( NF-κB) and cyclin D1 in osteosarcoma. Methods:Expressions of NF-κB p65 subunit and cyclin D1 in 46 cases of osteosarcoma, 15 of osteochondroma, and 5 of normal osseous tissue were determined by using immunohistochemistry method(SABC). The expressions of NF-κB in different histology grades and types and its relationship with cyclin D1 expression were determined. Results :Subunit of NF-κB p65 significantly expressed in osteosarcoma (67.4%), but was not found in ostechondroma and normal osseous tissue. Expression of NF-κB p65 has no relationship with osteosarcoma histology grades,and in high differentiation intraosseous group the expression was apparently higher than that in normal central group (P<0.05). Expression rate of cyclin D1 (73.9%) in osteosarcoma group was obviously higher than that in osteochondroma group (33.3%, P<0.05). Expression of NF-κB p65 had positive correlation with cyclin D1 protein in osteosarcoma (r=0.463, P< 0.01 ). Conclusion:NF-κB p65 was activated abnormally in osteosarcoma, and is closely correlated with the overexpression of cyclin D1 , which probably involves in the etiology of osteosarcoma by up-regulating cyclin D1 expression.
