aFGF对染铅大鼠海马神经元细胞浆ERK活性的保护作用

【摘要】 目的 :探讨酸性成纤维细胞生长因子 (aFGF)对染铅的原代培养大鼠脑神经元细胞外调节蛋白激酶(ERK)活性的影响。方法 :神经元培养 4~ 8d后 ,用 0 .6 μg/LaFGF及不同浓度铅处理细胞 ,用改良Takai法测定ERK活性。结果 :神经元染铅 30min ,在 0 .1~ 2 .0 μmol/L铅浓度范围内 ,ERK比活性明显升高 ,呈浓度依赖性 ,尤其 1.0~ 2 .0 μmol/L时最高。用aFGF预处理后再染铅时 ,在 1.0~ 5 .0 μmol/L时ERK比活性与对照组比较无明显差异。结论 :ERK比活性随着染铅浓度不同而发生改变。aFGF可保护神经元避免铅对ERK活性的影响。

【Abstract】 Objective: To observe the protective effect of acidic fibroblast growth factor(aFGF) on the activity of extracellular signal-regulated kinase( ERK) in primarily cultured hippocampal neurons of rats, which were exposed to lead. Methods: The hippocampal neurons of Wistar rats were primarily cultured for 4 to 8 days. Then the neurons were treated with aFGF and lead of different concentrations. The ERK activity in cytosal was determined by using improved Takai method. Results: From 0.1 to 2.0 μmol/L, lead activated ERK in dose-dependent manner. No significant difference of ERK activity in neurons was found between the groups when neurons was pretreated with aFGF. Conclusion: The activity of ERK in cytosal changed with the concentration of lead exposure. Exogenous aFGF can protect ERK activity in neuron from lead poisoning.
