尼莫地平微乳及自微乳的家兔口服生物利用度研究

【摘要】 目的 :以尼莫地平为模型药物 ,对其微乳、自微乳、片剂 3种制剂的生物利用度进行比较研究。方法 :新西兰大白兔单次口饲尼莫地平制剂 (2mg/kg) ,用HPLC法测定血浆中尼莫地平的浓度 ,根据血药浓度计算生物利用度。结果 :兔口饲尼莫地平后 ,血浓曲线均存在“双峰”现象。生物利用度从大到小依次为微乳、自微乳、片剂 ,且存在显著差异 (P <0 0 1)。结论 :微乳和自微乳能较大地提高尼莫地平兔口服生物利用度。

【Abstract】 AIM:To study the oral bioavailability of the microemulsion(ME) and self-microemulsion(SME) of nimodipine in rabbits METHOD: HPLC was used to determine the blood concentrations of nimodipine after the ME,SME and tablets of nimodipine were orally administrated to the rabbit respectively RESULT: “Double-peak” was observed in the blood concentration curve after each kind of preparation was orally administrated to the rabbit,and their bioavailabilities were ranked as follws:ME>SME>Tab The difference was significant(P<0 01) CONCLUSION:The results showed that the microemulsions and self\|microemulsions could markedly increase the oral absorption of the drug whereas the absorption was irregular
