高氡暴露地区居民肺癌组织已知基因和相关基因分析研究
Analysis of Some Tumor Known Genes and Related Genes-associated Genes in Lung Cancer Tissues of High Radon Exposure Residents
【摘要】 目的 研究高氡暴露地区居民肺癌组织中部分与肿瘤相关的基因突变规律和分离与肿瘤相关的新基因 ,旨在探讨肺癌发生的机制。方法 采用PCR扩增、PCR -SSCP电泳、mRNA差异显示和序列分析等方法。结果 5例标本p5 3PCR -SSCP电泳呈阳性结果 ,其中 3例经克隆测序发现有碱基置换。 7条差异片段经克隆测序后发现 ,其中NA7与GenbankEST中的A12 0 8667序列同源性达 95 %以上 ;NG2 与 5条基因序列同源性高达 98%;CA1可能是一个新的差异基因片段。结论 室内氡浓度在 2 0 0~ 3 3 8Bq·m-3 ·a-1范围内的 7例肺癌病例中有 5例发生p5 3基因突变 ,p5 3基因突变还可能与肺癌患者的年龄及吸烟史有关。从高氡暴露地区居民肺癌和正常肺组织中获得 3条与肿瘤相关的基因片段。
【Abstract】 Objective To analyze some tumor-associated genes in lung cancer tissues associated with residential radon exposure. Methods PCR amplification?PCR-SSCP electrophoresis?mRNA differentially expressed and sequence analysis. Results The five of 9 lung cancer cases were positive with p53 PCR-SSCP electrophoresis techniques,3 among the 5 were found bases substitution.One of which 7 sequences coded NA 7 had up to 95% of homology with A1208667 in EST of Genbank;other NG 2 had up to 98% of homology with five fragment sequences;CA 1 may be a new gene fragment. Conclusion The five of 7 lung cancer cases in 200-338 Bq·m -3 ·a -1 radon level had p53 gene mutation,p53 gene mutation may be related with age and smoking status.3 gene fragments were discovered from lung cancer and normal tissue of high radon exposure resident.
