黄精多糖对糖尿病鼠脑组织糖基化终产物受体mRNA表达的影响
Effect of Polygona-polysaccharose on Expression of RAGE mRNA in Brain of Diabetic Rats
【摘要】 目的 :探讨黄精多糖对糖尿病鼠脑组织糖基化终产物受体mRNA(RAGEmRNA )表达的调节作用。方法 :小鼠腹腔注射链脲佐菌素建立动物模型 ,采用逆转录聚合酶链反应法测定各组动物脑组织中RAGEmRNA的表达。结果 :糖尿病模型组脑组织RAGEmRNA的表达增加 ,其RAGE/β-actin值明显高于对照组 (P<0 01)。应用黄精多糖治疗后 ,治疗组脑组织RAGEmR NA表达较模型组降低 ,其RAGE/β-actin值显著低于模型组 (P<0 05)。结论 :黄精多糖能抑制糖尿病鼠脑组织RAGEmRNA的表达 ,对高血糖及糖基化终产物造成的脑组织损伤具有保护作用。
【Abstract】 OBJECTIVE:To study the regulative function of polygona-polysaccharose on the expression of RAGE mRNA in diabetic rats’brain.METHODS:Diabetic rat model was made by intraperitoneal injection of streptozotocin.RT-PCR method was used to examine the expression of RAGE mRNA in rats’brain.RESULTS:The expression of RAGE mRNA in brain tissue of diabetic rats was increased.The value of RAGE/β-actin in diabetic rats was significantly higher than that in the control group(P<0.01).After treatment with polygona-polysaccharose,the expression of RAGE mRNA in brain tissue was decreased in the treated group.The RAGE/β-actin value in the treated group was significantly lower than that in the model group(P<0.05).CONCLUSION:Polygona-polysaccharose may decrease the expression of RAGE mRNA in the brain tissue in diabetic rats and has protective effect on the tissue damage caused by hyperglycemia.
