高效液相色谱法测定红酵母细胞中的β-胡萝卜素
Deterimination of Beta-carotene in Red Yeast (Rhodotorula Glutinis) by High Performance Liquid Chormatography
【摘要】 采用反相高效液相色谱法(RP-HPLC)测定红酵母细胞中β-胡萝卜素的含量。发酵法得到的样品经过研磨,以丙酮-石油醚(1∶1 v/v)组成的溶剂提取β-胡萝卜素,经0.22μm(有机相)微孔过滤膜过滤净化,用RP-HPLC进行定量分析。色谱条件:分析柱为Hypersil RP-ODS C18(5μm,250mm×4.6mm i.d.);流动相:V(乙腈):V(四氢呋喃)=60∶40的溶液;检测波长451nm;以峰面积积分法定量。当β-胡萝卜素的质量浓度在2~18μg/mL范围时,其浓度与其峰面积之间有良好的线性关系(r=0.9971)。验证结果表明,该方法测定结果准确,能用来测定红酵母中的β-胡萝卜素。
【Abstract】 The sample of β-Carotene in red yeast (Rhodotorula glutinis) was determined by RP-HPLC with DAD/ UV detector at 451nm. The grinding samples obtained from fermentative method were extracted with acetone -petroleumether ( l : l V/V) followed by clean-up of the colander with 0.22μm microparous filter and quantitative analysis with RP-HPLC. The chromatographic conditions were as follows: RP-ODS C18 column, 250mm×4.6mm i.d., and mobile phase of acetonitrile-tetrahydrofuram (60:40, V/V), detection wave length: 451nm. The calibration curve showed a good linearity in 2-18μg/mL (r=0.9971) . The verified results demonstrated that this method is precise accurate and can be used for determination of β-carotene in red yeast.
