人下颌骨骨肉瘤细胞系HMOS-99的建立及形态学研究

【摘要】 目的:建立1株人下颌骨骨肉瘤细胞系,并观察其形态学特征。方法:骨肉瘤标本来自我院下颌骨骨肉瘤患者,裸小鼠皮下移植瘤传代11代,移植瘤组织块原代培养及连续传代培养。通过相差显微镜、微分干涉显微镜、HE染色、透射电镜、扫描电镜观察培养细胞的形态学特征。结果:HMOS-99细胞目前已传至95代,形态符合骨肉瘤细胞。结论:通过移植瘤方法建立了1株人下颌骨骨肉瘤细胞系HMOS-99,其形态学特征符合骨肉瘤细胞。

【Abstract】 PURPOSE: To establish a human mandibular osteosarcoma cell line by tumor tissue culture. METHODS: Tumor tissue of osteosarcoma of the jaw form one patient was transplanted to nude mice till the 11th passage. The tumor tissue was cultured in vitro in Ham F-12 medium with 10%FBS and 100U/ml penicillin and 100μg/ml streptomycin. The phase-control and differential interference microscopes, HE staining, TEM and SEM were used to observe the morphology of the cultured cells. RESULTS: The cells grew well and had passaged 95 generations. Morphylogical observations indicated that the cells were expected from the mesodermic origin of osteoblasts. CONCLUSIONS: The established cell line HMOS-99 has the morphological characteristics of osteosarcoma cell and will provide a good experiment model for the studies of osteosarcoma.
