5株O139群霍乱弧菌生物学性状研究

【摘要】 目的 研究从病人和水产品中分离到的 5株O139群霍乱弧菌的生物学性状。方法 参照《霍乱防治手册》对 5株菌的生长特征、生化特征、血清学反应及药物敏感性进行鉴定 ;应用多重PCR方法对所试菌株进行毒力相关基因检测 ,用Vero细胞测定方法检测ctxA阳性菌株霍乱肠毒素的表达。结果  5株O139群霍乱弧菌生物学性状和血清学反应与阳性对照菌体O139群霍乱弧菌MO4 5基本相同 ;2株来自重型病人和 1株来自水产品的菌株ctxA、zot、ace、tcpA、tcpI、hlyA、rtxA、rtxC、ompU、toxR 11种毒力相关基因全部阳性 ,经Vero细胞测定 ,其霍乱肠毒素效价达到 1∶16 0 ;在 2株来自轻型病人的菌株中仅hlyA、rtxA、rtxC、toxR 4种毒力相关基因为阳性。 结论 O139群霍乱弧菌致病 (泻 )的决定基因簇并非仅为ctxA、zot、ace、cep等毒力相关基因。

【Abstract】 Objective To investigate the biological characteristics of five vibrio O139 strains isolated from patients and seafood.Methods The basic biological characteristics,including growth properties,biochemical characteristics,serological responses,drug sensitivities,were detected according to the routine methods recommended by“A manual of prevention and treatment for cholera”.Multiplex PCR was used to detect the toxin genes,including ctxA、zot、ace、tcpA、tcpI、hlyA、rtxA、rtxC、ompU、toxR. The vero cell cytotoxicty assay was applied in testing the level of CT production of ctxA + strains.Results The biological characteristics and serological responses of 5 strains were similar to positive control strain Mo45.In 2 strains isolated from the heavy patients and 1 strain isolated from seafood.All of 11 toxin genes were found in the 3 strains,of which CT titers reached 1∶160.In the other 2 strains isolated from light patients only 4 toxin genes were found.Conclusion In the determinant of pathogenicity related gene in vibrio cholerae O139 strains there were others genes besides ctxA、ace、cep、zot toxin genes.
