大豆异黄酮对去卵巢大鼠骨形成蛋白的影响

【摘要】 目的 探讨大豆异黄酮和钙对去卵巢大鼠无机盐、骨形成蛋白 2 (BMP2 )和转移生长因子 - β1(TGF - β1)的影响。方法 取 2 8只雌性SD大鼠随机分为 4组 :假手术组、去卵巢组、去卵巢补钙组〔5 0mg/ (kg·d)〕和去卵巢加大豆异黄酮组〔大豆异黄酮 10 0mg/ (kg·d)〕 ,10周后测骨密度、血清和尿液钙、钠、钾含量。用免疫组化法测骨骼中BMP2 和TGF - β1的表达。结果 去卵巢组大鼠血清和尿钙、钠含量显著增高 ,而骨密度、血清和尿钾及骨BMP2 和TGF - β1表达降低 ;去卵巢加大豆异黄酮后大鼠血清钙和尿钙、钠显著降低 ,骨密度和骨BMP2 、TGF - β1显著性升高(P <0 0 5或P <0 0 1)。单纯补钙组与去卵巢组相比只有TGF - β1有显著性差异 (P <0 0 5 )。结论 大豆异黄酮可调节去卵巢大鼠无机盐代谢 ,促进BMP2 和TGF - β1表达 ,预防去卵巢大鼠的骨质疏松

【Abstract】 Objective To study the effect of soybean isoflavone and calcium on mineral and the expression of bone BMP\-2,TGF-β 1 in ovariectomized rats.Methods Twenty eight female SD rats were randomly divided into four groups:sham,ovariectomized,ovariectomized added calcium〔50?mg/(kg·d)〕,ovariectomized added soybean isoflavone〔100?mg/(kg·d)〕.The content of serum and urine calcium,sodium,potassium were determined after ten weeks.The Streptavidin-Peroxidase Histostain TM -Plus Kits was used to detect bone BMP\-2 and TGF-β 1 expression.Results The calcium,sodium of serum and urine in ovariectomized group increased significantly.The bone mineral density,and potassium of serum and urine,bone BMP\-2 and TGF-β 1 decreased significantly.The serum calcium and urine calcium,sodium in ovariectomized added soybean isoflavone group decreased significantly,but the bone mineral density and bone BMP\-2 and TGF-β 1 expression increased significantly( P <0 05 or P <0 01).Only the TGF-β 1 expression in ovariectomized added calcium group had significantly increace than that of ovariectomized group( P <0 05).Conclusion The soybean isoflavone may adjust the mineral metabolism and prompt the bone BMP\-2,TGF-β 1 expression to prevent the osteoporosis of ovariectomized rats.
