γ-干扰素基因转染对宫颈癌细胞株细胞周期及表面抗原的影响
The impact of cell period and cell surface antigen in cervical cancer cell lines by interferon-γ gene transfer
【摘要】 目的 :研究IFN γ基因转染对宫颈癌细胞表面抗原、细胞周期的影响 ,初步探讨IFN γ基因治疗抗肿瘤作用的机制。方法 :制备含人IFN γ基因的真核表达质粒pcDNA 3 IFN γ ,脂质体包浆后转染人宫颈癌HeLa细胞株 ,流式细胞仪检测基因转染后HeLa细胞表面HLA DR抗原表达、细胞凋亡及细胞周期的变化。结果 :IFN γ基因转染后 ,HeLa细胞表面的HLA DR的表达量明显增强 (P <0 .0 1)。转染IFN γ基因促进了HeLa细胞的凋亡 (P <0 .0 5 ) ,细胞的S期比例随作用时间延长而呈逐渐降低趋势 ,显示了基因转染后DNA的合成代谢受到抑制。结论 :IFN γ基因转染可有效增强宫颈癌细胞表面抗原的表达 ,诱导机体产生抗肿瘤免疫应答 ;并可能通过抑制细胞DNA的合成 ,促进细胞的凋亡 ,抑制肿瘤细胞的增殖等机制发挥抗肿瘤作用
【Abstract】 Purpose:To explore the tumor cell responses after IFN-γ gene transfer by analyzing the expression of cell surface antigen,apoptosis and the impact of cell period. Methods:We constructed IFN-γ gene expression vector-pcDNA3- IFN-γ, and transfected it into HeLa cell lines by Lipofectamine. The expression of IFN-γ?the Human Leukocyte antigen(HLA-DR) after gene transfer were measured. The apoptosis of tumor cells and the variety of cell period were also measured to investigate the anti-tumor mechanism by IFN-γ gene therapy. Results:The results showed that the HLA-DR expression in Hela cell lines increased significantly(P<0.01) after IFN-γ gene transfer, and the IFN-γ gene transfer can effectively induce the apoptosis of tumor cells, the proportion of DNA synthesis period decreased gradually after IFN-γ gene transfer. This infer that the synthesis of DNA can be repressed and the proliferation of tumor cells be inhibited. Conclusions:The cell surface antigens can be effectively increased by IFN-γ gene therapy, which induces strong antitumor immunity. The other possible anti-tumor mechannisms may include repression of the synthesis of DNA, induction of apoptosis of tumor cells and inhibition of the proliferation of tumor cells.
