EGCG对人乳腺癌MDA-MB-435细胞株细胞增殖的抑制及机制
Inhibition effects of epigallocatechin-3-gallste (EGCG) on cell cycle of hum an breast cancer cells MDA-MB-435 and its mechanism
【摘要】 目的 :研究绿茶提取物表没食子儿茶素 3 没食子酸酯 (epigallocatechin 3gallste ,EGCG)对人乳腺癌细胞株细胞周期的影响及机制。方法 :利用细胞增殖测定试剂盒 (CellProliferationAssaykit)来制作细胞生长曲线 ;流式细胞仪分析EGCG作用于人乳腺癌MDA MB 43 5细胞株前后细胞周期的变化 ;RT PCR及Western印迹法研究肿瘤细胞株加药干预前后细胞周期抑制因子 p2 1waf1/cip1的mRNA及蛋白表达的变化。结果 :EGCG可明显抑制乳腺癌细胞MDA -MB -43 5细胞的增殖活性 ,40 μg/mlEGCG干预后 ,MDA MB43 5细胞主要阻滞于G0 /G1期 ,在2 4小时阻滞最为明显。在 48及 72小时也有阻滞。对照组G0 /G1期的比例 49.92 % ,40 μg/mlEGCG干预 2 4、48及 72小时后G0 /G1期的比例分别为 71.56%、67.2 0 %和 61.59%。EGCG 40 μg/ml处理肿瘤细胞株细胞后 2 4小时可检测 p2 1mRNA和蛋白表达提高。 结论 :绿茶有效成分可抑制乳腺癌细胞的增殖 ,这可能与其诱导 p2 1表达从而抑制细胞周期的时相转换有关 ,这表明绿茶可能在乳腺癌的治疗上具有一定的前景
【Abstract】 Purpose:To study the effect of EGCG(extracts fr om green tea) on cell cycle in human breast cancer cell line and its mechanism. Methods:Cell proliferation assay kit was used to produce the dose-response curve. Flow cytometric assay was used to evaluate the cell cycle changes on MDA-MB435 cells with and without EGCG. The expression of protein was detected by Western blot. Results:EGCG could inhibit the proliferation of the breast canc er cells MDA-MB-435, 40 μg/ml EGCG induced G 0 /G 1 phase arrest and the entrance to S phase. Both mRNA and protein level of p21 waf1/cip1 were up regulated in MDA-MB435 cells when treated by 40μg/ml EGCG. Conclus ions:Green tea can inhibit the growth of human breast cancer cells, perhaps by means of p21 and therefore inhibiting the progression of the cell cycle.
