H2O2对PC12细胞par-4和NF-κBP65基因表达的影响
Effects of H2O2 on expression of the PC12 cell gene par-4 and NF-κBP65
【摘要】 目的 探讨 H2 O2 对 PC12细胞前列腺细胞应答凋亡蛋白 - 4 ( prostate apoptosis response- 4 ,par- 4 )和核转录因子 - κBP6 5 ( NF- κBP6 5 )基因表达的影响。方法 用不同终浓度 H2 O2 ( 0~ 4 0 0 nm ol/ L)处理 PC12细胞 8h,或终浓度 2 0 0 nm ol/ L H2 O2 处理 PC12细胞不同时间 ( 0~ 8h) ,采用 RT- PCR检测 par- 4、NF- κBP6 5 m RNA表达变化 ,Western blot检测 Par- 4和 NF-κBP6 5蛋白表达的变化。结果 随 H2 O2 浓度从 10 0~ 4 0 0 nmol/ L增加 ,par- 4、NF-κBP6 5 m RNA表达和 Par- 4、NF-κBP6 5蛋白表达水平逐渐增加 ;2 0 0 nm ol/ L H2 O2 作用 PC12细胞 0~ 8h,par- 4 m RNA表达和 Par- 4蛋白表达 ,在 2~ 8h随时间延长表达增加 ;在 0~ 8h NF- κB P6 5 m RNA表达和 NF- κBP6 5蛋白表达无明显变化。结论 H2 O2 可时间、剂量依赖性的增加 par- 4 m RNA表达和 Par- 4蛋白表达 ;H2 O2 可剂量依赖性的增加NF-κBP6 5 m RNA表达和 NF-κBP6 5蛋白表达 ;NF-κBP6 5 m RNA表达和 NF-κBP6 5蛋白表达无时间依赖性
【Abstract】 Objective To determine the effects of H 2O 2 on the expression of gene par-4 and NF-κB in PC12 cells.Methods After the PC12 cells were treated with H 2O 2 [0~400nmol/L] for 8h,or treated with200nmol/L H 2O 2 0 to 8 hours,mRNA expression of par-4 and NF-κBP65 weredetected by RT-PCR and protein expression of par-4 and NF-κBP65 were detected by Western-blot.Results The mRNA and protein expression of par-4 and NF-κBP65 gene were upregulated with concentration of H 2O 2 [100~400nmol/L];After the PC12 cells were treated with 200nmol/L H 2O 2 0 to 8 hours,the mRNA and protein expression of par-4 increased from 2 to8 hours and 4 to 8 hours respectively,while those of NF-κBP65 had no obvious change.Conclusions H 2O 2 increased the mRNA and protein expression of par-4 gene in a time and dose-dependent manner;The mRNA and proteinexpression of NF-κBP65 gene were upregulated by H 2O 2 in a dose-dependentmanner,but not in a time-dependent manner.
