大孔吸附树脂分离纯化葛根与山楂叶中总黄酮的研究
Separation and purification of total flavone from Kudzuvine root and hawthorn leaves by macroporous resin
【摘要】 目的 :研究大孔吸附树脂分离纯化葛根与山楂叶中总黄酮的工艺条件。方法 :采用紫外分光光度法测定混合物中总黄酮的含量。结果 :D10 1型树脂对混合物中总黄酮有良好的吸附 ,其吸附分离的工艺条件为葛根黄酮饱和吸附量10 .10mg·g-1,山楂叶黄酮饱和吸附量 11.2 8mg·g-1,吸附流速 3mL·min-1,洗脱剂为 80 %乙醇 ,洗脱剂用量为 2 0倍药材量 ,洗脱流速 3mL·min-1。结论 :D10 1大孔吸附树脂可用作葛根与山楂叶中总黄酮的精制
【Abstract】 AIM: To set up the method for adsorbing and separating total flavone in Radix Puerariae and in Folium Crataegi. By macroreticular resin. METHODS: UV Spectrophotometry was used to analyze the content of total flavone in mixture. RESULTS: The D101 was the best for adsorbing and separating total flavone in the following technological conditions: the maximum adsorbing capacity for flavone in Radix Puerariae was 10.10mg·g -1; the maximum adsorbing capacity for flavone in Folium crataegi was 11.28mg·g -1; the current velocity was 3ml·min -1, the eluting reagent 80% ethanol (20 times the volume of medicinal herbs) and the eluting velocity 3ml·min -1. CONCLUSION: D101 resin could be used to refine the total flavone in Radix Puerariae and in the Folium Crataegi.
