维生素E、硒对急性肝损伤期肝细胞凋亡影响的研究

【摘要】 目的: 了解饲料维生素E (VE) 和硒 (Se) 对大鼠急性损伤期肝细胞凋亡的调节作用。方法: 采用腹腔内注射50%CCl4制备大鼠急性肝损伤模型,在饲料中添加适量VE(250 mg/kg)和Se(0.2 mg/kg)进行营养干预,用ALT、AST等肝功能指标及病理组织学观察的方法检测抗氧化剂对肝细胞的保护作用;用原位凋亡(TUNEL)技术结合组织学形态观察的方法检测抗氧化营养素对肝细胞凋亡的影响。结果: 注射CCl4后大鼠ALT和AST值均显著增高;肝细胞凋亡数少于正常对照组; 饲料补充VE和Se后肝损伤程度减轻;肝细胞凋亡数增高。结论: 饲料中添加适量VE和Se可减轻CCl4对大鼠肝细胞的损伤并促进其凋亡。

【Abstract】 Objective: Effects of dietary supplementation of vitamin E and selenium on apoptosis of degenarated hepatocytes were investigated in the rat model of acute liver damage induced by intraperitoneal injection with CCl4. Methods:Hepatocyte apoptosis was determined by terminal deoxynucleotidyl transferase UTP nick end labeling (TUNEL) technique. Results:Apoptotic hepatocyte in both pathological group and intervention group was less than in normal control group, meanwhile, apoptotic hepatocytes in intervention group was more than in pathological group at the same time point. Conclusion:Dietary supplementation of proper vitamin E and selenium can promote the apoptosis of degenarative hepatocytes in acute liver damage.
