表面等离子体共振法研究抗生物素抗体与生物素作用
Study of Interaction Between Biotin and Its Monoclonal Antibody by Surface Plasma Resonance(SPR)
【摘要】 在有些情况下 ,抗生物素抗体被用来替代亲和素的作用。采用基于表面等离子体共振 (SPR)原理的生物分子相互作用分析技术研究了抗生物素抗体与生物素相互作用的动力学特征。结果表明 ,抗生物素抗体与生物素的结合速率常数为 2 5 9× 10 4L/ (mol·s) ,解离速率常数为 3 77× 10 -3 s-1,结合常数为 6 86×10 6L/mol。
【Abstract】 Avidin and biotin are widely used in biology due to the high affinity between them. In some cases anti-biotin antibodies are used in stead of avidin. In this paper, the kinetic characters of anti-biotin antibody and biotinylated bovine serum albumin(BSA) were studied by biomolecular interaction analysis(BIA) technique based on surface plasma resonance(SPR). The association rate constant, dissociation rate constant and affinity constant between anti-biotin antibody and biotinylated BSA were found to be 2.59× 10 4 L/(mol·s), 3.77×10 -3 s -1 and 6.86× 10 6 L/mol, respectively.
