谷氨酰胺对鲑鱼胚胎(CHSE)细胞生长和代谢的影响

【摘要】 鉴于鱼类和哺乳动物的谷氨酰胺代谢途径有所不同 ,为了优化CHSE细胞的培养基和大量培养的过程控制 ,考察了谷氨酰胺对CHSE细胞生长和代谢的影响 .在CHSE细胞批培养中 ,无谷氨酰胺的M199培养基可以支持CHSE细胞的生长 ,当初始谷氨酰胺浓度c =0 .5 4mmolL-1时 ,细胞可达到最高密度nmax/ 10 5mL-1=16 .19;而更高浓度的谷氨酰胺 (c =1.76~ 5 .6 2mmolL-1)对细胞生长有抑制作用 .在c=0~ 5 .6 2mmolL-1初始谷氨酰胺浓度的批培养中 ,葡萄糖的利用和乳酸的生成基本一致 ,随谷氨酰胺浓度的升高 ,Qglc和Qlac增加 ,Yn/ glc降低 .另外 ,随着初始谷氨酰胺浓度的增加 (c=0~ 3.33mmolL-1) ,谷氨酰胺的消耗增加 ,Yn/ gln和Yammo/ gln分别下降 5 8%和 5 0 % ;当初始谷氨酰胺浓度继续增加时 ,谷氨酰胺的消耗、Yn/gln和Yammo/gln基本不变 .此外 ,当初始谷氨酰胺浓度为 0时 ,丙氨酸成为消耗性的氨基酸 ;Yala/gln先随初始谷氨酰胺浓度的增加 (c=0 .5 4~ 1.76mmolL-1)而增加 ,而后又随初始谷氨酰胺浓度的增加 (c=1.76~ 3.33mmolL-1)而降低 ,最后维持不变 .图 4表 3参 16

【Abstract】 The difference has been known in glutamine metabolism between fish and mammalian. In order to optimize the medium and the culture process for CHSE cells, the effects of glutamine were investigated on the cell growth and metabolism. Glutamine-free medium-199 could support the growth of CHSE cells, as well as medium-199 with glutamine. The maximum cell density, 16.19×105 mL -1, was obtained at 288 h in the culture with 0.54 mmol L -1 glutamine. However, higher glutamine (c= 1.76~ 5.62 mmol L -1) inhibited the growth of CHSE cells. With the increase of initial glutamine from 0~5.62 mmol L -1, the total utilization of glucose and the production of lactate were almost not affected.But the yield coefficient of cell to glucose (Y n/glc) was reduced, and the specific rates of glucose consumption (Q glc) and lactate production (Q lac) increased. On the other hand, with the increase of c gln (0~3.33 mmol L -1), the glutamine consumption was increased,the yield coefficient of cell to glutamine (Y n/gln) and ammonia to glutamine (Y ammo/gln) dropped by 58% and 50%, respectively, and then kept constant with c gln up to 5.62 mmol L -1. Besides, alanine was consumed in the absence of glutamine, and the yield coefficient of alanine to glutamine (Y ala/gln) first increased with the increase of c gln from 0.54 mmol L -1 to 1.76 mmol L -1, then dropped with c gln up to 3.33 mmol L -1, and finally kept constant between 3.33 mmol L -1 and 5.62 mmol L -1 c gln.
