喜树愈伤组织培养及喜树碱的产生研究

【摘要】 目的:研究影响喜树愈伤组织生长和喜树碱产生的培养条件。方法:选用不同培养基对喜树愈伤组织诱导和继代培养的基础上,进行细胞悬浮培养,分化出胚状体,用薄层层析法检测其是否含有喜树碱。结果:最适合培养基为B5基本培养基,附加激素为1.0 mg·L-1萘乙酸及0.2 mg·L-1激动素。结论:利用组织培养的方法分化出的胚状体与喜树碱生物合成有一定的相关性。

【Abstract】 Objective: To study the condition influenced on the growth of callus of compothecine acuminata decaisne and the production of camptothecin. Method: Based on the various culture medium used in inducing and sub-cuhure of camptotheca callus, cells were cultivated in suspension, the embryoid was differentiated and thin-layer chromatogram was applied to detect the camptothecin. Results: The most suitable culture medium was B5, and the additional growth regulators were 1.0 mg·L-1 NAA and KT 0.1 mg·L-1. Conclusion: There was correlation between the embryoid differentiated by tissue cuture and the biosynthesis of camptothecin.
