高压芒刺静电场对小鼠胸腺细胞和腹腔巨噬细胞中CD2、CD48表达的影响

【摘要】 目的:通过时程/量效研究观察不同强度芒刺高压静电场对表面分子CD2、CD48表达的影响。材料和方法:采用不同强度芒刺高压静电场对小鼠进行全身辐射,荧光免疫流式细胞术检测小鼠胸腺细胞CD2蛋白和腹腔巨噬细胞CD48蛋白表达的变化。结果:剂量—效应结果显示,在10kV~20kV之间处理组的CD2、CD48表达量显著高于对照组(P<0.01);在30kV以上电压表达量明显低于对照组(P<0.01);15kV电压时间进程研究结果表明,CD2于处理后2h表达开始增加,在4h~8h表达量达到高峰,与对照组比较差异显著(P<0.001),24h时表达量趋于恢复,接近对照组;CD48于处理后4h开始增加,高峰出现在8h,与对照组比较差异显著(P<0.001),24h时的表达量接近对照组;35kV电压时间进程研究结果表明,CD2于处理后2h表达量即达到最低值,之后表达量逐步回升,但持续到24h未能恢复到对照组水平;CD48表达量的下降晚于CD2,在4h开始下降,最低值出现在处理后8h,其后逐步回升。结论:不同强度高压芒刺静电场可引起CD2、CD48表达量不同的生物效应,二者的相互作用在免疫调节反应中发挥重要功能。

【Abstract】 Objective: To observe the changes in the expression of CD48 on peritoneal macrophages and CD2 on thymocytes after different doses of high voltage prickle electrostatic field by time-course and dose-effect studies. Material and Methods: Whole body of mice was radiated by high voltage prickle electrostatic field. Immuno-fluorescent flow cytometry(FCM) was adopted to detect the changes of CD48 and CD2 expressions. Results: The dose-effect studies showed that CD48 and CD2 expressions increased significantly after 10 kV~20 kV of high voltage prickle electrostatic field(P<0.01), while 30 kV and above decreased the expressions of CD48 and CD2(P<0.01); Time-course results of 15 kV showed that CD2 expression on thymocytes increased beginning from 2 h and reaching its peak at 4 h~8 h after high voltage prickle electrostatic field(P<0.001), while CD48 expression increased from 4 h and reached its peak at 8 h after 15 kV of high voltage prickle electrostatic field(P<0.001); Time-course results of 35 kV showed that 35 kV of high voltage prickle electrostatic field decreased CD2 expression from 2 h and then recovered gradually. Simultaneously, CD48 expression on peritoneal macrophages decreased beginning from 4 h and reaching its minimum at 8 h(P<0.001). Conclusion: The different doses of high voltage prickle electrostatic field might cause different changes of CD48 on peritoneal macrophages and CD2 on thymocytes,which suggested that high voltage prickle electrostatic field had potential immune-regulatory effects and the changes of CD48 and CD2 might play important roles in immune modulation.
