【摘要】 目的 :调查新疆维吾尔自治区 (新疆 )静脉注射毒品者中输血传播病毒 (TTV)的感染状况及基因型别。方法 :采用巢式PCR技术检测 10 2例汉族、维吾尔族、回族静脉注射毒品者血清中TTVDNA ,以 38例正常体检者为对照 ,比较静脉注射毒品者中TTVDNA阳性率是否与共用注射器有关。将一例维吾尔族TTV阳性扩增产物插入pGEM -TEasy载体 ,测序并序列分析。结果 :静脉注射毒品者TTVDNA阳性率为 32 35 % ,显著高于正常体检者 (5 2 6 % ) ,P <0 0 5。静脉注射毒品者中汉族、维吾尔族、回族TTVDNA阳性率分别为 32 6 1% (15 4 6 )、35 14 % (13 37) %、2 6 32 % (5 19) ,各民族之间差异无显著性 (P >0 0 5 )。静脉注射毒品者TTVDNA阳性率与共用注射器有关 (P <0 0 5 )。一例维吾尔族TTVDNA部分核苷酸测序 ,与日本株 (AB0 0 8394 )比较同源性为 98% ,属于G1a型。结论 :静脉注射毒品者是TTV感染的高危人群。一例维吾尔族TTV核酸序列与日本株 (AB0 0 8394 )相比有高度同源性 ,属于同一基因型别更多还原

【Abstract】 Objective: To study the prevalence and homology analysis of transfusion transmitted virus (TTV) in intravenous drug abusers (IVDUs) in Xinjiang.Methods: Using Nest-PCR, the TTV DNA was detected in 102 IVDUs (including Han, Uygur and Hui) and 38 healthy subjects. The relationship of TTV DNA infection was analyzed between the IVDUs who shared injectors and those who used their own. A TTV DNA fragment was amplified from Uygur IVDUs group and was cloned into pGEM-T Easy plasmid, and then sequenced. The sequences were analyzed by GenBank/Blast biosoftware. Results: Of 102 IVDU S, 33 were found TTV positive. The positive rate was 32 35% in IVDUs group, which was much higher than that in healthy subjects(5 26%), P<0 05. In IVDUs group, the TTV positive rate showed no statistically significant difference among Han group (32 61%, 15/46), Uygur group (35 14%, 13/37) and Hui group (26 32%, 5/19), P>0 05, but a significant difference existed between those who shared injectors and who not (P<0 05) in the IVDUs group. The TTV DNA sequence and homology analysis indicated 98% homology to that of the Japan strain (AB008394), belonging to G1a genotype.Conclusion: The IVDUs were high-risk group of TTV in Han, Uygur and Hui in Xinjiang. The TTV sequence, which amplified from Uygur IVDUs, has a striking homology to reported Japan strain (AB008394) and belongs to the same genotype.更多还原
